Pyrrosia is a kind of traditional Chinese herbs,recorded in several Classics of traditional Chinese Herbal medicine.It has the efficacy of relieving stranguria by diuresis,benefiting yin and nourishing lung,and hemostasis.This paper studied and clarified the chemical composition of Pyrrosia,which provides an important information for the development of Pyrrosia herbs.Pyrrosia petiolosa(christ)ching(one of the prescribed sources in the Chinese Pharmacopoeia,planted in Northeast,Northwest,Southwest China,and the middle and lower reaches of the Yangtze River.Due to its rich natural resources and mature cultivation technology,it is the most widely used in the market nowadays,so we collected the Pyrrosia petiolosa(christ)ching as the sample.The studies achieved the following results: 200 kg Pyrrosia medicinal material was crushed and extracted with 70% ethanol under reflux.The extract was extracted with dichloromethane,ethyl acetate and n-butanol in this order.The extract was separated and purified by column chromatography methods such as silica gel column chromatography,Sephadex LH-20,RP-C18,pre-HPLC,etc.37 compounds were isolated from the nbutanol fraction of Pyrrosia.According to the physical and chemical properties of the compound and MS,NMR and other spectral data were used to identify the structure.The chemical structure of each compound was determined as follows:(±)Eriodictyol-7-O-?-D-pyrannoglucuronide Methyl Ester(1),chlorogenic acid methyl ester(2),(+)-Catechin(3),2-O-?-Glucopyranosyl-5-methoxy-benzoic acid methyl ester(4),vanillic acid(5),protocatechuic acid(6),esculetin(7),epicatechin(8),4-caffeoylquinic acid methyl ester(9),4?-carboxymethyl-(-)-epicatechin methyl ester(10),caffeic acid(11),P-coumaricacid-4-O-?-D-glucopyranoside(12),caffeic acid-4-O-?-D-glucopyranoside(13),Eriodictyol-7-O-?-glucoside(14),maesopsin-6-O-glucopyranoside(15),3?,5?,5,7-tetrahydroxy-6-C-?-D-glucopyranosyl-flavanone(16),isolariciresinol-6-O-?-D-glucopyranoside(17),3-(?-D-Glucopyranosyloxy methyl)-2-(4-hydroxy-3-methoxyphenyl)-5-(3-hydroxypropyl)-7-methoxydihydrobenzofuran(18),isorhamnetin-3-O-?-D-glucoside(19),kaempferol-3-(2-acetylglucopyranose)-7-arabinofuranoside(20),kaempferol-3-O-?-D-glucoside(21),kaempferol-3-O-?-L-rhamnoside(22),kaempferol-7-O-?-L-arabinoside(23),7,8-dihydro-9?-hydroxyl-3?-methoxyl-8-hydroxymethyl-7-(4-hydroxy-3-methoxyphenyl)-1?-benzofuranpropanol 9?-O-?-D-glucopyranoside(24),kaempferol-3-O-?-L-rhamnose-7-O-?-L-arabinoside(25),alphitonin-4-O-glucoside(26),Eriodictyol-6-C-?-D-glucoside(27),(7S,8R)dihydrodehydrodiconiferyl alcohol 4-O-?-D-glucopyranoside(28),catechin-3-O-?-L-rhamnoside(29),Dihydrocedarin(30),3-O-feruloylsucrose(31),alangioside A(32),kaempferol-3-O-?-D-glucose-7-O-?-L-arabinoside(33),kaempferol-3-O-?-D-glucose-7-O-?-Dcelery glycoside(34),kaempferol-3-O-?-D-xylpyranose-7-O-?-L-arabinofuranoside(35),(±)hesperetin-7-O-?-D-glucopyranoside(36),erythro-7,9,9'-trihydroxy-3,3'-dimethoxy-8-O-4'-neolignan-4-O-?-D-glucopyranoside(37).In the studies,37 compounds were isolated and identified from Pyrrosia,29 compounds were firstly isolated and identified in the species of Pyrrosia petiolosa(Christ)Ching.Except for compounds 10,13,16,the remaining 26 compounds 4,6-9,12,15,17-20,22-32,34-37 were isolated from Pyrrosia plants for the first time.There were 17 flavonoids,5 lignans(15,17,18,24,28,37),4 phenolic acids and other compouds.This issue studies and clarifies the main chemical composition of Pyrrosia,provided information for the research on the pharmacological active substances of the Pyrrosia,and also established foundation for the quality evaluation of Pyrrosia.
