Objective:This study is to investigate the therapeutic effect of Pyrrosia herbs.on adjuvant arthritis(AA)in rats and its mechanisms.Method:1.80 KM male mice were randomly divided into 8 groups:model group, dexamethasone group, pyrrosia lingua and pyrrosia mollis (high doses, middledoses, low doses) groups.The drugs were administrated by intragastric gavage once daily respectively, after 7 days treatment. The inflammatory models induced by Croton oil (auricle smear method)and Carrageenan (injecetion under the aponeurosis)respectively, to explore the effct of pyrrosia lingua and pyrrosia mollis on the moused’s ear edema and hine paw edema.80 KM female mice were randomly divided into 8 groups:control group, aspirin group,pyrrosia lingua and pyrrosia mollis (high doses, middle doses, low doses) groups. The drugs were administrated by intragastric gavage once daily respectively, after 7 days treatment. The pain models were established by acetic acid and hot plate in mice, to explore the effct of pyrrosia lingua and pyrrosia mollis on the pain threshold and twister number.2.90 Vistar male rats were randomly divided into 9 groups, physiological saline group,model group, tripterygium wilfordiipplycoside group, pyrrosia lingua and pyrrosia mollis (high doses, middledoses, low doses) groups Complete Freund ’s adjuvant was used to induce A in rats. Secondary paw swelling of AA rats was measured with volume meter. The levels of IL-10, TNF-α, PGE2 on the serum were estimated by enzyme linked immunosorbent assay(ELISA), and the levels of IL-1 β,TNF-α, PGE2 in the fluid within joints were estimated radioimmunoassay.We detected superoxide dismutase(SOD),total antioxidative capacity(T-AOC) activity and malonaldehyde(MDA) in serum by special kits to assess the antioxidant effects of samples on AA.Results:1. Anti-inflammatory experimental resuts showed that,compared with the model group, the moused’s ear edema and hine paw edema in the pyrrosia lingua and pyrrosia mollis high doses and middle doses groups were decreased(P<0.01 or P<0.05). Compared with the pyrrosia mollis group, the moused’s hine paw edema in the pyrrosia lingua high doses and middle doses groups were decreased significantly(P<0.01); Analgesic experimental resuts showed that,compared with the control group, the pain threshold in the pyrrosia lingua and pyrrosia mollis high doses and middle doses groups were increased (P<0.01 or P<0.05), the twister number of mice in the pyrrosia lingua and pyrrosia mollis high doses and middle doses and low doses groups were decreased (P<0.01 or P<0.05). Compared with the pyrrosia mollis group, the pain threshold in the pyrrosia lingua high doses and middle doses groups were increased significantly(P<0.01), the twister number of mice were decreased without significant difference(P>0.05).1.pyrrosia lingua and pyrrosia mollis could significantly inhibit primary acute inflammatory swelling and secondary inflammatory reaction in rats(P<0.05 or P<0.01)3. The serum IL-β,TNF-α,PGE2 and MDA levels of the treatment groups were lower than those of the model group, the SOD and T-AOC levels were developed, and two samples showed the inhibition on the serum IL-10,TNF-α,PGE2 and MDA expression in rat adjuvant arthritis, and improved the ability of antioxidation. Pyrrosia lingua was more effective than Pyrrosia mollis.4. The joint fluid IL-1β,TNF-α,PGE2 levels of the treatment groups were lower than the model group,and Pyrrosia lingua was more effective than Pyrrosia mollis.5.The white blood cells, neutrophils, lymphocytes, monocytes,eosinophils of AA rats increased significantly compared with the control group, mean corpuscular volume, mean hemoglobin were lower than control group, Pyrrosia lingua and Pyrrosia mollis can decreased hite blood cells, neutrophils, lymphocytes, monocytes, eosinophils and increased mean corpuscular volume, mean hemoglobin, Pyrrosia lingua was more effective than Pyrrosia mollis.Conclusion:Pyrrosia lingua and Pyrrosia mollis show a certain role in the treatment of rheumatoid arthritis,and can effectively control the rat metatarsal swelling, reduce the content of serum IIL-1, TNF- alpha and PGE2, maintain the relative content of serum SOD, T-AOC, inhibit MDA content in serum, reduce lipid peroxidation damage, thereby preventing the articular cartilage from breaking, and decrease the pain. In the experimental process, no adverse reaction are seen. Pyrrosia lingua and Pyrrosia mollis are effective for the treatment of rheumatoid arthritis, and Shi Wei show a stronger effect.
