Objective: Gastric cancer(GC)is the fifth most common cancer in the world,and it has the third highest mortality rate.The prognosis of patients with gastric cancer is closely related to the stage and metastasis of the tumor.C-reactive protein(CRP)is an acute phase protein synthesized by the liver,CRP was originally used to monitor patients with sepsis and determine their prognosis.In recent years,many studies have shown that plasma CRP is generally elevated in tumor patients,and plasma CRP levels are closely related to tumor pathological stage and prognosis,however,the current research is only based on clinical statistical research,and the related research results are controversial.Expression of CRP in gastric cancer tissues and cell lines in vitro and whether it affects gastric cancer cells in vitro is unclear.The purpose of this experiment is to use immunohistochemical techniques to study the expression and distribution of CRP in gastric cancer and adjacent cells.To investigate whether plasma high CRP in gastric cancer patients is partially derived from tumor tissue,and to further explore the malignant biological behavior of CRP on gastric cancer cells in vitro.Methods: This topic is divided into two parts: Part I: The expression and distribution of CRP in cancer tissues and adjacent tissues of patients with gastric cancer Using immunohistochemical techniques,34 cases of gastric cancer and adjacent tissue samples were collected,to observe the expression of CRP in gastric cancer and adjacent tissues.Part 2: Expression of CRP in gastric cancer cell lines and its effect on the biological behavior of gastric cancer cells1.Using polymerase chain reaction technology to amplify and construct the CRP lentivirus overexpression vector.Using immunofluorescence and immunoblot tests,To observe its overexpression.2.Using Polymerase Chain Reaction technology,testing the expression of CRP in AGS,HGC-27,NCI-N87 and MKN-45 cell lines,and select the appropriate cells as experimental cells.3.Using the overexpression virus to infect the target cells,CCK8、Transwell migration and invasion test were used to investigate the effect of overexpression of CRP on the malignant biological behavior of the target cells.Results: 1.Immunohistochemistry showed that the expression of CRP in gastric cancer was higher than that in adjacent tissues.2.CRP was generally expressed in gastric cancer cells,AGS,HGC-27,NCI-N87 and MKN-45.The expression abundance of CRP in AGS and NCI-N87 cells was medium,but it was low in HGC-27 and MKN-45 cells.So we selected HGC-27 cells as the experimental cell.3.The expression of CRP in HGC-27 cells increased significantly after infection with lentivirus vector.4.When the target cells were infected with over expressed lentivirus vector,CRP was over expressed in gastric cancer cells.This makes hgc-27 cell proliferation,migration and other malignant biological behavior ability increase.Conclusions: The expression of CRP in gastric cancer is higher than that in adjacent tissues,and CRP is generally expressed in gastric cancer cells.In this experiment,we constructed a CRP overexpression lentivirus vector,and infected hgc-27 cells with overexpression lentivirus.The results showed that the expression level of CRP was significantly increased after virus infection,and the value-added,invasion and migration ability of HGC-27 cells increased.Therefore,raising CRP level in HGC-27 cells can promote the malignant biological behavior of cells.
