| Objective:To explore the expression and significance of autophagy-related factors microtubule associated protein 1 light chain3B(LC3B),Beclin1 and p62 in oral lichen planus lesions.Methods:1.41 cases of OLP lesions(20 cases of non-erosive OLP,21 cases of erosive OLP)and 15 cases of normal oral mucosal as the control group.Immunohistochemical techniques were used to detect the expression and distribution of LC3B,Beclin1 and p62 proteins in OLP lesions and Reverse transcriptase quantitative PCR(RT-q PCR)was used to detect the expression levels of LC3B,Beclin1 and p62 m RNA in OLP lesions.The relationships between the above three indicators and clinical pathological features were analyzed.2.16 cases of OLP lesions(8 cases of non-erosive OLP,8 cases of erosive OLP)and 15 cases of normal oral mucosa.The expression of LC3B,Beclin1,p62 protein and LC3B-II/LC3B-I ratio were detected by Western blot(WB).The relationship between LC3B-II/LC3B-I ratio and clinical pathological features of OLP was analyzed.Results:1.The positive expression rates of LC3B and p62 proteins in OLP lesions were 68.3%(28/41)and 58.5%(24/41)respectively,which were higher than those in the control group 33.3%(5/15),20%(3/15)(P<0.05).The positive expression rates of LC3B and p62 proteins in erosive OLP group were higher than those in non-erosive OLP(c12=4.498,P1=0.034;χ22=5.528,P2=0.019)and the positive expression rate of Beclin1 protein in OLP lesions was 19.5%(8/41),which was lower than that in control group 47%(7/15),but the positive expression rate of Beclin1 protein was no statistically significant between the non-erosive OLP and erosive OLP(P>0.05);2.The levels of LC3B and p62 m RNA expression in OLP lesions were higher than those in the control group(Z1=-4.561,P1<0.001;Z2=-4.783,P2<0.001)and LC3B,p62 m RNA expression level in the erosive OLP group were higher than those in the non-erosive OLP(Z1=-2.869,P1=0.004;Z2=-2.947,P2=0.003),As not the same as these two factors,Beclin1 m RNA expression level in OLP group was lower than that in the control group(Z=-2.433,P=0.015),but the difference between the two OLP types was not statistically significant(P>0.05).3.The ratio of LC3B-II/LC3B-I in OLP group was higher than that in the control group(t=-2.447,P=0.021)and the ratio in non-erosive OLP was higher than that in erosive OLP(t=-2.378,P=0.032).Spearman correlation analysis showed that the LC3B-II/LC3B-I ratio was negatively correlated with OLP clinical typing and basal cell liquefaction degeneration(r1=-0.569,P1=0.021;r2=-0.537,P2=0.032).Conclusions:1.The expression of autophagy-related factors LC3B,Beclin1 and p62 in OLP lesion tissues is abnormal,which is related to OLP clinical typing.It is suggested that autophagy-related factors may play a role in the local immune mechanism of OLP lesions.2.The autophagy level of OLP local tissues is elevated,and it is related to the clinical type and the degree of basal cell liquefaction degeneration.It is suggested that the autophagy level of erosive OLP is relatively low,and this may be one of the reasons of the increased damage in OLP lesions. |
