Objective Determination of renal hypertensive rats and non renal hypertensive rats group changes of intestinal bacteria diversity,Bifidobacterium and Lactobacillus content,explore the relationship between intestinal flora and renal hypertension,for the treatment of renal hypertension to provide new research ideas and reference.Methods 8 week old male Sprague-Dawley rats were randomly divided into 2K1C model group(n=8)and sham operation group(n=8),Adaptive feeding week and RVH model was prepared by 2K1C surgery(The middle of the left renal artery was a silver clip with a diameter of 0.2mm and the right renal artery was not treated).The systolic pressure of tail artery was measured by non-invasive tail cuff method.At 4th week after the operation,intestinal feces were collected and the bacteria DNA was extracted from the feces.The diversity of intestinal flora in2K1C rats was analyzed by PCR-denaturing gradient gel electrophoresis.The dominant bacteria(bifidobacterium and lactobacillus)in the intestinal tract were quantitatively detectd by real-time fluorescence quantitative PCR.Results 4 weeks after modeling,Compared with the sham operation group,systolic pressure of tail artery in 2K1C model group was significantly higher than that in sham operation group(164.77 mm Hg vs 122.08 mm Hg,F=38.178,P<0.001).However,there was no significant change in the systolic pressure of tail artery in sham operation group compared with the previous model(F=26.864,p<0.001;F=21.568,p<0.001).In the 2K1C model group,the diversity of intestinal flora in the two groups was lower than that in the sham operation group.The similarity between the two groups was low,and the similarity index was 0.29.2K1C of Bifidobacterium model group rat intestinal flora in Bacillus(2.335 x 10-3ng/g)and Lactobacillus(0.048 ng/g)was significantly lower than that in sham group(1.805 x 10-2ng/g,0.139 ng/g),the difference was statistically significant(2.335×10-3ng/g vs 1.805×10-2ng/g,0.048 ng/g vs 0.139 ng/g,P<0.05).Conclusion Renal hypertension rats with renovascular hypertensive rats compared to intestinal flora diversity decreased,Bifidobacterium and Lactobacillus decreased significantly,suggesting that the intestinal flora may be involved in the occurrence and development of renal hypertensive rats.
