The problem of food safety has become the focus of the public attentions.Beta-agonist(β-agonist,commonly known as "Clenbuterol")as veterinary drugs from food of animal,is one of the most serious chemical residue hazards since they have the highest detected rate.Developing a method that is high-throughput,high sensitiveness and high speed to detect the p-agonist has become a key link ofβ-agonist regulation.Electrochemical immunosensor is combined with the method of electrochemical and immunosensor analysis technology,and it has broad application prospects in food,environment and other fields.Appling the excellent electrochemical properties of graphene and the broad specificity of multi-epitope-antibody to electrochemical analysis.In this paper some new types of electrochemical immunosensors for the beta-agonist analysis are developed.The electrochemical immunosensor was successfully applied to the detection of beta-agonist in real samples.Chapter 1:An overview of the current situation and the main detection method ofβ-agonist in food,the applications and research status of electrochemical immunosensor are summarized in this chapter.We introduce the advantages of graphene is applied to electrochemical immunosensor,the background and main research content of this paper.Chapter 2:BSA-CL-SAL antigen was prepared,and anti-BSA-CL-SAL antibody was prepared by animal immune.The graphene was prepared by oxidation reduction reaction.We developed two kinds of electrochemical immunosensor of graphene/chitosan/CL and graphene/chitosan/SAL,which are successfully applied to the analysis of standard solution of clenbuterol,ractopamine,salbutamol,terbutaline,mabutero and toybbutero.The results show that the sensor has a wide linear range and a low detection limit.Using this method,the immunosenor of graphene/chitosan/SAL was applied to the analysis of clenbuterol in real samples including lean meat,fat and liver of pig,the range of recovery rate was 82.6%-109.9%.The immunosenor achieved detection of β-agonist with high sensitiveness and multi-residue.Chapter 3:Anti-BSA-SAL-RAC antibody was prepared by animal immune based on BSA-SAL-RAC antigen.The graphene was prepared by oxidation reduction reaction.We developed the electrochemical immunosensor of graphene/chitosan/SAL,which is successfully applied to the analysis of standard solution of clenbuterol,salbutamol,terbutaline,and the results show broad linear range and high high sensitiveness.The method is applied to the analysis of clenbuterol in real samples include lean meat,fat and liver of pig,and the results show satisfying recovery rate which range from 84.7%to 106%.Chapter 4:BSA-SAL-RAC antigen and Anti-BSA-SAL-RAC antibody were prepared by animal immune.We use graphene prepared liquid-phase exfoliation,and developed three kinds of electrochemical immunosensors of graphene/CL,graphene/SAL and graphene/SAL,which is successfuLly applied to the analysis of standard solution of clenbuterol,ractopamine,salbutamol,terbutaline,mabutero and toybbutero.By using this method,the immunosenor of graphene/EAC is applied to the analysis of clenbuterol and ractopamine in real samples including lean meat,fat and liver of pig,the resuLts quite fit the bill that the recovery rates was 80.5%~106.0%and 87.6%~103.3%.Electrochemical immunosensor of multi-residual detection was prepared in this paper,it has good stability and high sensitiveness.Furthermore,the success of the strategy employed in this work provides a rapid and effective method for the detection of residual p-agonist in food. |
