Study On The Extraction, Microcapsule Preparation And Biological Activity Of Hericium | | Posted on:2017-01-21 | Degree:Master | Type:Thesis | | Country:China | Candidate:L N Yao | Full Text:PDF | | GTID:2431330512963752 | Subject:Food processing and safety | | Abstract/Summary: | PDF Full Text Request | | Hericium erinaceus is commonly used for medicine or food has a variety of biological activities.This research adopted the microwave extraction method to extract hericium ericium from hericium erinaceus polysaccharide extraction residue materials,in order to improve the utilization rate of hericium erinaceus.Using the single factor and orthogonal experiments to optimize the formulation:when the microwave treatment was conducted at 60℃for 25 min,the microwave power was700 W and the liquid to solid ratio was 1:20,under these conditions,the Hericium erinaceus extracts yield reached 4.28%.In this paper,Hericium erinaceus extracts microcapsules were prepared byβ-cyclodextrin assisted by ultrasonic and using PLGA as wall material by emulsification solvent evaporation technique.The single factor was used to optimize the formulation according to the efficiency of encapsulation.The results demonstrated that Emulsification-solvent evaporation method was developed to prepare PLGA-Hericium erinaceus-MS with higher entrapment efficiency.Using a scanning electron microscope to observe appearance shape,the result is that hericium erinaceus/PLGA microcapsules has smaller particle size and the embedding effect is better.The influence of formulation and manufacture was studied by the single factor experiments,then,the optimal formulation was verified by response surface methodology:the optimum prescription was Hericium erinaceus:PLGA=1:1.64(w/w),15%(w/v)PLGA dichloromethane solution,stirring speed is 1200 r/min,and the encapsulation efficiency was 99.66%.Then the physical and chemical properties of the microcapsule products were investigated:Fourier transform infrared spectroscopy show that the embedding process of microcapsule is a physical mixture.The characteristics of drug release were determined in vitro and the results showed that microcapsules have good embedding effects and preparations could induce sustained release of Hericium erinaceus.Finally,the antioxidant activities of microspheres and the effect on nerve growth factor(NGF)release from rat cerebral cortical astrocytes were studied.The results showed that microcapsules had a certain antioxidant activity.In the ranges of the experiment,there was a linear relationship between the reducing power and concentration of microspheres,the IC50 of removing DPPH·and ABTS·+free radicals were 7.63 mg/mL and 10.28 mg/mL,respectively.Microspheres can enhance NGF release from rat cerebral cortical astrocytes.On the concentration of 2.5μg/mL,the promote of microspheres for the secretion of radial nerve cells of rats nerve growth factor concentration was 69.33±0.73 pg/mL. | | Keywords/Search Tags: | Hericium erinaceus, Microspheres, Antioxidant activity, Nerve growth factor | PDF Full Text Request | Related items |
