| Objective:Through observing the amountof Collagen fibers,a-SMA and FN,which is influenced by the four QingGuangAn effective elements and QingGuangAn granules,in the filtration canal scarring process after glaucoma surgery,to discuss that the mechanism of the process of filtration canal scarring.Methods:Select48 NewZealand long ears white rabbits(weight from 1.5 to 2 kilogram),divide them into eight groups with six rabbits each group using SPSS 19.0.These groups are:A group(blank group),B group(model group),C group(MMC group),D group(group with first efficacious element),E group(group with second efficacious element),F group(group with third efficacious element),G group(group with fourth efficacious element),H group(group with QingGuangAn suspension).However,A group would be deal with nothing,while B,C,D,E,F,and G groups would do with trabeculectomy and iris root resection surgery,in addition,C group should also use MMC along with its surgery.After surgeries,give all the rabbits medicine in the first day,and messure their intraocular pressure in the next day,the seventh day,the fourteenth day and the day 28.Simultaneously,observed the anterior segment of eyes,filtration bleb and the eyes’ general states.Four weeks later after giving medicine,all rabbits were executed by air embolism,get their whole organization of filtration canal,dye with Masson,and then observe the expression of collagen fibers in filtration blebs area.Detect the expressions of α-SMA and FN in filtration blebs area with immunohistochemistry.Results:The IPO of C,E,H groups’ rabbits rise very slowly.It is unobvious of the IPO’s rising up to the fourth week,and the comparison with other groups show that the difference is statistically significant.C,E,H group could maintain effective filtration blebs,while the blebs of B group gradually changed to be nonfunctional and the blebs disappear up to the fourth week.D,F,G group could maintain a few blebs in the fourth week after surgery.In the comparison of the area ratio of collagen fibers between each groups,B,D,G group’s ratio is bigger than 0.05,the difference is not statistically significant.However,the ratio difference between other groups is statistically significant.Comparing with theα-SMA expressions,P>0.05 between B group and G group,which say the difference is not statistically significant,while for all other groups,the difference is statistically significant.Comparing with the FN expressions,P>0.05 between B group,D group and G group,which say the difference is not statistically significant,while for all other groups,the difference is statistically significant.Conclusions:(1)The production of filtration canal scar is strong related with theCell proliferation of collagen fibers,myofibroblasts and fibroblast after Glaucomasurgery(2)MMC,QingGuangAn suspension and the second effective element of QingGuangAn are all effectiveto the inhibition of cell proliferation of α-SMA,FN and collagen fibers,which means they are able to anti-scarring.(3)Through inhibiting the cell proliferation ofα-SMA,FN and collagen fibers,MMC,QingGuangAn suspension and the second effective element of QingGuangAn could inhibit the cells of myofibroblasts and fibroblast. |
