Objective: To establish an SD rat acute soft tissue injury model with effects were compared to investigate the efficacy and possible therapeutic mechanism of Danggui-San in the treatment of acute soft tissue injury in SD rats,and to provide a theoretical basis for the drug in the treatment of acute soft tissue injury.Methods: Eight-week-old male Sprague-Dawley rats(SD rats)were selected and weighed 268.81±31.9 g after 7 days of adaptive feeding.Randomly divided into three groups according to the random number table method,namely saline group(n=24),Qili-San group(n=24)and Danggui-San group(n=24)were used to manufacture models using heavy object combat method.After modeling,each group of rats was randomly divided into 4 groups(n=6),i.e.group a.(Day 1),group b(day 3),group c(day 7),group d(day 12).After modeling,the normal saline group was given an equal amount of physiological saline,and the drug control was fed with Qili-San suspension.The experimental group was fed with Danggui-San suspension.At each time point,the body weights of rats in each group were weighed;the skin temperature was measured;the change of soft tissue injury index was observed;the changes of histomorphology and PGE2 content were detected by optical microscope;the experimental data were analyzed by SPSS19.0 The software performs statistical analysis and processing.Results:(1)the difference of bilateral skin temperature: the three groups were compared with each other.There was no significant difference between the two groups in the same phase point 22(P > 0.05),which had better balance.In the first to third days after the model,the body weight of the rats in the saline group,the seven LP group and the Danggui San group decreased slightly,the body weight of the saline group was the largest and the seven in the group was the least,but the weight loss of the three groups was not significantly different(P > 0.05).After the third day,the weight of the three rats began to rise continuously,The physiological saline group was more obvious,the two groups of drug groups were close to the number of twelfth days,the three groups of rats had little difference in weight,and there was no significant difference between the 4 heavy objects and to use the clinical experience of Prof.Zheng Huaixian,a famous expert in military medicine in our hospital,to treat Danggui San,as an oral therapeutic agent,and to observe and record bilateral rats periodically.The skin temperature difference in lower extremities,soft tissue injury symptoms,histomorphology,and prostaglandin E2(PGE2)changes in local tissues,and the traditional prescription for the treatment of traumatic injuries in traditional Chinese medicine orthopedics The curative no significant difference between the two groups(P > 0.05).(3)tissue section observation: the histomorphological score of the three groups on the first day was compared,no significant difference(P>0.05),third days,seven in the scattered group,the angelica powder group and the saline group compared,the difference was significant,the difference was significant,there was no significant difference(P>0.05)in the Danggui San group and the seven Li group(P>0.05);seventh days,Compared with the normal saline group,the difference was significant(P<0.05).There was no significant difference between the normal saline group and the seven Li group(P>0.05),and the twelfth day,the group of Danggui San was P<0.05 compared with the seven Li group and the saline group,and the difference was statistically significant.The seven Li group and the physiological saline group were statistically significant.There was no significant difference in the group(P>0.05).(4)PGE2 content measurement: first days,22 of the three groups had no significant difference(P>0.05);third,seventh,Twelfth days,seven len group,angelica powder group and saline group compared with the saline group,P<0.01,there was significant difference,there was no significant difference between the two groups(P>0.05),no statistical significance.Conclusion:Danggui San can effectively improve the local congestion,tissue swelling and activity disorders after acute soft tissue injury;it has the function of reducing the content of PGE2 in injured soft tissue,probably by reducing the concentration of PGE2 in the local damaged tissue to eliminate inflammation,inhibit pain and antipyretic effect.Danggui San has a good effect on the treatment of acute soft tissue injury.Moreover,the Danggui San scattered group has a simple,cheap and good taste,and it is worth promoting in clinical practice.
