Objective: To investigate thre effects of total flavonoids from Cyclocarya paliurus on proliferation and differentiation in 3T3-L1 preadipocytes,and thre effects on glucose consumption,free fatty acid(FFA)production in IR 3T3-L1 adipocytes.And thren explore mechanisms of drugs by measuring gene expression of Glut4 mRNA,IRS-1mRNA and PPAR? mRNA.Methods: 3T3-L1 preadipocytes were cultured and treated with Cyclocarya paliurus extractive of different concentration at each time,while blank control group(Complete medium),medium group(1 ‰ DMSO),positive drug(Met,2 ?g/mL)were set also,the proliferation ratio was detected by MTT assay.3T3-L1 preadipocytes were induced to be differentiated into 3T3-L1 adipocytes,the differentiation was observed by oil red O staining.Then 3T3-L1 adipocytes models of insulin resistance were established successfully and treated with drugs,control group and model group were added.Glucose consumption was assayed by glucose oxidase method(GOD-POD)and free fatty acid(FFA)level by colorimetric method.The gene expression of Glut4 mRNA,IRS-1 mRNA and PPAR? mRNA was detected via reverse transcriptase PCR(RT-PCR)analysis.Results: 1.Effects on proliferation and differentiated of 3T3-L1 preadipocytes.Compared with medium group,Met promoted proliferation of 3T3-L1 preadipocytes significantly(P<0.01),total flavonoids from Cyclocarya paliurus promoted proliferation of 3T3-L1 preadipocytes significantly(P<0.01);Compared with medium group,Met promoted differentiation of 3T3-L1 preadipocytes significantly(P<0.01).Total flavonoids from Cyclocarya paliurus promoted differentiation of 3T3-L1 preadipocytes significantly(P<0.01).2.Effect on glucose consumption and FFA production of 3T3-L1 IR adipocytes.Whether in basic or insulin stimulated state,compared with IR medium group,Met improved glucose uptake in 3T3-L1 IR adipocytes significantly(P<0.01).Total flavonoids from Cyclocarya paliurus improved glucose uptake significantly(P<0.01).Compared with IR medium group,Met inhibited FFA production significantly in 3T3-L1 IR adipocytes significantly(P<0.01).total flavonoids from Cyclocarya paliurus inhibited FFA production significantly(P<0.01).3.Effects on gene expression of GLUT4 m RNA,IRS-1 mRNA and PPAR? mRNA in 3T3-L1 IR adipocytes.Compared with IR model group,Met up-regulated gene expression of PPAR? mRNA in 3T3-L1 IR adipocytes significantly(P<0.01).total flavonoids from Cyclocarya paliurus up-regulated gene expression of PPAR? mRNA in 3T3-L1 IR adipocytes significantly(P<0.01).Whereas had no significant on Glut4 mRNA and IRS-1 m RNA.Conclusion: 1.The total flavonoids from Cyclocarya paliurus promoted the proliferation of 3T3-L1 preadipocytes and inhibited their differentiation significantly.total flavonoids from Cyclocarya paliurus promoted the glucose consumption of IR adipocytes and inhibit the production of FFA,improved the metabolism of glucose and lipid significantly.2.In conclusion,total flavonoids from Cyclocarya paliurus can improve the insulin resistance in 3T3-L1 adipocytes with a dose-dependent manner.The mechanism may be that significantly up-regulated gene expression of PPAR? mRNA in IR adipocytes to improve the insulin signaling pathway.
