Cyclocarya paliurus(Batal.) Iljinskaja as a special health food rescources in china,which can improve hyperglycemia and hypertension, have great potential for development.Research works of Cyclocarya paliurus mainly focused on the separating and extracting of active ingredient and evaluating the biological efficacy. There are few researchers,research the mechanism of how Cyclocarya paliurus active ingredients work. In this article, we use water extract-alcohol precipitation method to obtain Cyclocarya paliurus crude polysaccharide from Cyclocarya paliurus dry leaves, and evaluate the function of Cyclocarya paliurus crude polysaccharides on diabetic mice. Separating and purifying Cyclocarya paliurus flavone and polysaccharide by column chromatography and alcohol precipitating, dialysis, magnesium oxide decoloring method. Studing the effects of Cyclocarya paliurus polysaccharide(CPP) and Cyclocarya paliurus flavone(CPF) on glucose consumption of insulin resistance Hep G2 cell and activity of alpha-glucosidase and the capability of oral glucose tolerance in hyperglycemia mice. The main results are as follows:1. Separating and purifying Cyclocarya paliurus flavone and polysaccharide via column chromatography and alcohol precipitating, dialysis, magnesium oxide decoloring method from dry leaves. The content of polysaccharide is 66.31%, tested by sulfuric acid-anthrone method; the content of flavone is 80.50%, tested by sodium nitrite- aluminum nitrate- sodium hydroxide colorimetry method.2. The crude Cyclocarya paliurus polysaccharide(CRP) was obtained by water extracting-alcohol precipitating from dry leaves powders. Through multiple intraperitoneal injection of low concentration streptozotocin(40 mg/kg) and feeding with high-fat diet established hyperglycemia mice model. These mice were randomly divided into normal control, hyperglycemia model control, positive control, CRP low-dose, CRP moderate-dose, and CRP high-dose groups(10 mice per group). After 35 days administration, the capability of oral glucose tolerance was measured. The value of oral glucose tolerance area under curve(AUC) of positive control group and CRP low, medium and high dose groups was significantly lower(p<0.05 or p<0.01) than model control group,and the respective proportion of decline were 24.74%, 18.97%, 29.36% and 22.15%.3. Induced Hep G2 cell with high concentration insulin(10 ?g/m L) to establish insulin resistance cell model and divided model cell into Cyclocarya paliurus polysaccharide(CPP), Cyclocarya paliurus flavone(CPF) and dimethylbiguanide groups.After 24 h administration, used glucose assay fit to measure the gross glucose consumption of cells(?GC), detected glucose consumption of unite cell(?GC/OD) by MTT assay.Compared with model control group, the value of ?GC was significantly increased(p<0.01), and the rate of increase were 621.69%, 173.49%, 234.94%, 339.76%, 96.39%,239.76%, 279.52%.4. The inhibitory activity on alpha-glucosidase of CPP and CPF was evaluated,compared with acarbose, in vitro. The inhibition ratio of alpha-glucosidase of acarbose(1.00 mg/L), CPP(0.13?0.25?0.50?1.00?2.00?4.00 mg/m L) and CPF(0.13?0.25?0.50?1.00?2.00?4.00 mg/m L) were 47.52%, 43.95%, 45.46%, 35.62%, 42.28%, 43.16%,44.82%, 35.78%, 33.53%, 35.91%, 37.46%, 46.17%, 86.00%.Experimental results show that by using column chromatography and alcohol precipitating, dialysis, and magnesium oxide decoloring method can obtain high purity of CPF and CPP. Crude Cyclocarya paliurus polysaccharide extracts can significantly improve the capability of oral glucose tolerance. CPF and CPP both can significantly increase the consumption of glucose and improve the insulin resistance in IR-Hep G2 cell.CPP and CPF with different concentration have inhibitory effect on alpha-glucosidase in vitro. The inhibitory ability of CPP is not related with the concentration of CPP, but the inhibitory ability of CPF is positively correlated with the concentration of CPF. Those indicated that CPF and CPP have effects on increasing the glucose consumption of cell and decreasing the activity of alpha-glucosidase, thus alleviating hyperglycemia.
