Stem cells have strong differentiation ability,which can continue to self-renewal and multi-directional differentiation into body’s various tissues and organs.a large number of bone marrow stem cells can differentiate into various cell types,mainly including hematopoietic stem cells(hematopoietic stem cells,HSCs),another kind is bone marrow mesenchymal stem cells(mesenchymal stem cells,MSCs)。Bone marrow mesenchymal stem cells derived from mesoderm,under the right conditions to grow into liver cells.Bone marrow mesenchymal stem cells are a kind of ideal experimental study of allograft transplantation for the treatment of cirrhosis of the liver of engineering seed cells,can benefit for the treatment of chronic liver disease.In the specific environment,bone marrow mesenchymal stem cells can promote the repair of liver injury,improve liver function.Basic and clinical research at home and abroad has confirmed between bone marrow mesenchymal stem cell transplantation to the disease such as cirrhosis,has a definite therapeutic effect.At present,in many countries has been carried out between the autologous bone marrow mesenchymal stem cells for clinical treatment of liver cirrhosis,treatment result is more optimistic,and found no obvious side effects.Although it has the therapeutic effect of a variety of diseases,the security of its clinical application is still in the test,and many mechanism is unclear.there are three main kinds of transplantation among treatment of end-stage liver disease,which respectively is the way of portal vein,hepatic artery way and caudal vein approach.In order to simulate the clinical characteristics of stem cell transplantation in the treatment of liver cirrhosis,we made the experiment by intraperitoneal injection of carbon tetrachloride and olive oil mixture together and alcohol feeding method of liver cirrhosis in rats model,and then use fluorescent CM-Di I mark between in vitro cultured rat bone marrow mesenchymal stem cells in three ways,and then transplanted bone marrow MSCs into model rats,comparison of liver cirrhosis in rats after three transplantation organization form,the number of stem cell homing,evaluate the degree of liver function,fibrosis index,stem cell transplantation in the treatment of end-stage liver disease for clinical finding the best transplantation way to survey after transplantation of cells homing distribution and confirmation after transplantation in the case of a change of diseased tissue provide the basis of experimental research.Methods:1.buy SD school-age rats,then extracted BMSCs using the method of whole bone marrow adherent bone marrow and separation.Culture to the third generation of bone marrow MSCs and using CM-Di I labeled MSCs.2.established cirrhosis rats model by using carbon tetrachloride by intraperitoneal injection of liquid,mixture of olive oil and alcohol feeding method,increasing the concentration of carbon tetrachloride and wine.HE and Masson staining in liver tissue,observe the degree of liver cirrhosis.By automatic biochemical analyzer test serum albumin,cereal third transaminase,aspertate aminotransferase,direct bilirubin,indirect bilirubin,etc.3.via the portal vein,hepatic artery way and caudal vein approach transplantation BMSCs marked into cirrhotic model in rats.4 weeks after transplantation of liver tissue for HE and Masson staining,observe the degree of liver cirrhosis.Caudal vein transplantation in the rat serum,by automatic biochemical analyzer test serum albumin,cereal third transaminase,aspertate aminotransferase,direct bilirubin,indirect bilirubin,etc.Results:1.laser confocal microscope CM-Di I tag MSCs in intrahepatic distribution by three ways BMSCs transplantation and 4 weeks after the homing no obvious difference was found between the number of MSCs in the liver.2.changes in liver histology and pathology results(1)4 weeks after BMSCs transplantation,observe the liver gross appearance,the degree of cirrhosis rats transplantation group by three ways were eased;Cirrhosis of the liver transplantation group no significant change.(2)HE staining and Masson staining showed after transplantation groups decrease fiber bundle in the rat liver biopsy of liver tissue,and reduced necrosis.the number of false flocculus decreased,alleviate the damage of liver cells,the degree of cirrhosis of the liver have improved.But in three different ways transplantation group of liver pathology to improve the degree of no significant difference.3.serologic test resultsThrough three ways transplanted BMSCs to cirrhotic model in rats,4 weeks later found model rat liver transplantation group were obviously improved,serum albumin is increased significantly of the cirrhosis of the liver transplantation group(P<0.05),transaminase,bilirubin is decreased significantly of the cirrhosis of the liver transplantation group(P<0.05).Conclusion:by three ways Hepatic artery,portal vein and caudal vein approach transplantation BMSCs liver cirrhosis model of rat treatment,after 4 weeks test the number of BMSCs homing in liver no statistical difference,the Degree of liver fibrosis and liver function improvement;Three ways to transplant stem cells to treat liver cirrhosis model of rat are effective,and the effect is similar.
