Object:Sambucus chinensis Lindl is a significant effect of herb medicine in folk used to treat traumatic injury and fracture injury.Abundant resources and widely distributed.Domestic and foreign research is relatively weak,and it has not been recorded in "Chinese Pharmacopoeia and local standards in Guizhou",which hinders its further development and utilization.Based on the literature review and the pre-experiment,the target components were separated and identified by phytochemical method.On the basis of obtaining the target composition,the multi-index components of the herbs were established,and the fingerprints and total polyphenol content determination method,and then the Guizhou origin of 15 batches of grass to analyze.Include Water,ash(total ash and acid insoluble ash),leachate and lead,cadmium,mercury,arsenic,copper and 9 kinds of organochlorine,in accordance with the technical guidelines of the Chinese Pharmacopoeia.Pesticide residues and other related items of detection.This study enriched the basic research of the national medicine in Guizhou province and provided a scientific basis for promoting the quality standards and the quality control of related products by analyzing the internal quality.Method:1.Based on the Guizhou resource survey,the collection of herbal medicine is representative and based on "Chinese Flora","Chinese Materia Medica"and other experts and experts with qualified qualifications for the identification of varieties of resources.2.Based on the pre-experiment,the components of the herbs were separated by silica gel column chromatography and preparative high performance liquid chromatography(HPLC),and the structures were identified by means of nuclear magnetic resonance(NMR);3.The HPLC method and UV method were used to establish the simultaneous determination of 3 indexes and the determination of total polyphenol content.4.The HPLC fingerprints of 15 batches of different herbs were established,and the common peaks were identified,and the common patterns were established.The similarity of each batch was calculated.The differences between the batches were found by cluster analysis and principal component analysis.5.According to the requirements of "Chinese Pharmacopoeia" to improve the quality standards,including identification of traits,microscopic identification,thin layer identification,water,total ash,acid insoluble ash,heavy metals,organic pesticide residues and taste,Usage usage,precautions and storage,and to develop drafting instructions.Results:1.15 batches of elderberry samples from different habitats of Guizhou were collected and identified,and the dried whole plants were identified by Sambucus chinensis Lindl.2.The structure of the two compounds separated by fixation kaempferol-3-O-β-D-(6-O-acetyl-glucopyranose)-7-O-β-D-glucopyranoside and kae-mpferol-3-O-β-D-(2-O-acetyl-glucopyranose)-7-O-β-D-gluco-pyranoside.3.The two isomers JGC-1 and JGC-2 were isolated by chemical constituents and the total contents of chlorogenic acid,JGC-1 and JGC-2 from 0.11 to 3.37 mg·g-1.There was some difference in the content of total polyphenol of S chinensis from different habitats and different organs.The content sequence of different organs of the same plant are as follows,leaf>whole grass>stem>root.Antioxidative activity test showed the sample result.The contents of total hydroxybenzene from different habitats were significantly different and leaves had the relatively higher antioxidant activity.4.3 common peaks were identified by the HPLC fingerprint of Sambucus chinensis.The similarity of the 15 batches of Sambucus chinensis 0.88-0.99,the quality of the collected samples was generally consistent,higher similarity,because of differences in bone growth environment,leading to the similarity between the samples are still large differences.5.The main characteristics of the identification and microscopic identification of the Sambucus were established.Established TLC methods of S chinensis with chlorogenic acid and kaempferol as active ingredient.The results showed the water content Was from 5.93%to 8.32%,the total ash was from 6.54%to 10.61%,the water-soluble extract was from 17.84%to 27.43%,the ethanol-soluble extract was from 15.53%to 23.62%,the mercury content Was from 0.062 mg·Kg-1 to 0.101 mg·Kg-1 the arsenic content was from 0.039 to 1.364 mg·Kg-1,the copper content Was from 3.502 to 10.500 mg·Kg-1,the copper content Was from 0.073 to 0.414 mg·Kg-1,the copper content Was from 4.102 to 14.750 mg·Kg-1,9 organochlorine pesticide residues were not exceeded,the chlorogenic acid concent was from 0.04%to 0.46%.6.Drafting and established the quality standard of Sambucus Chinese.Conclusion:1.The structure of the two compounds separated by fixation kaempferol-3-O-β-D-(6-O-acetyl-glucopyranose)-7-O-β-D-glucopyranoside and kae-mpferol-3-O-β-D-(2-O-acetyl-glucopyranose)-7-O-β-D-gluco-pyranoside.2.The method of simultaneous determination of multi-index,fingerprint and total polyphenol content was established.The quality of elderberry was evaluated from the system and macroscopic point of view.On this basis,the quality standard of Chinese herbal medicine was established,Thus laying a foundation for promoting the industrialization of elder grass in Guizhou.
