This dissertation is composed of two parts:literature review and experimental research.The part of literature review:The first literature review:Research progress of microRNA-155 in psoriasisThe second literature review two:Progress in treatment of psoriasis with traditional Chinese medicine monomerOBJECTIVE:1.reveals the expression characteristics and localization of microRNA-155 in psoriasis,and clarifies whether it plays an important role through SOCS1/STAT3 pathway,and can be a therapeutic target.2.This paper expounds whether and how Paeonol is used to control psoriasis by microRNA-155 regulation of related signal transduction molecules and inflammatory factors,and provides objective experimental basis for the clinical application of salvianolic phenol from Target and target.METHODS:After shaving the back of the BALB/c mice,the mouse model of psoriasis was induced by applying Imiquimod cream in the back of the mice.The mouse model of psoriasis induced by midazolam was used by Tripterygium,MicroRNA-155 Simulator,MicroRNA-155 inhibitor,Paeonol,MicroRNA-155 inhibitor+Paeonol.The changes of skin lesions in the back of each group were compared with the PASI scores,and the contents of IL-17,IL-23,IFN-γ,SOCS1 and STAT3 in serum were detected by ELISA.The expression of the above indexes in the skin lesions of each group was detected by RT-PCR method.Results:It was found that the skin of BALB/c mice coated with imidazole Mott Cream showed thickening of skin,invasive erythema and scales,and some of them dispersed in the bleeding point.Combined with the results of he staining,it is suggested that the molding is successful.The back skin lesions of tripterygium,Paeonol,MicroRNA-155 inhibitor,MicroRNA-155 inhibitor+Paeonol group were improved in different degree than that of the model group,and the MicroRNA-155 Simulator group was significantly aggravated.The PCR method showed that the IL-17,IL-23,INF-γ,SOCS1 and STAT3 in the skin of the model group were significantly higher than that in the blank group,and the expression of the other groups decreased obviously,and the MicroRNA-155 Simulator group increased obviously.Conclusion:Experiments show that microRNA-155 may be one of the targets for the treatment of psoriasis,elucidate the mechanism of the action of Salvianolic phenol in the treatment of psoriasis,and provide guidance and direction for the clinical and scientific research of TCM monomer in the treatment of psoriasis.
