Research background and objective:The proportion of male infertility is about 15% to 50% among the infertile couples. In recent years, as environmental degradation, social competition increase and other affection factors, the incidence rate showes an increasing trend. In contrast, the pathogenesis of male infertility is still currently unclear, and therefore there is still no targeted and effective drugs in Western medicine neither at home nor abroad. Now the drugs of clinical treatment of male infertility are androgen, dopamine receptor agonists, receptor blockers,anti-oxidants and so on. In rigorous double-blind controlled trial, a considerable number of drugs are only equal to placebo;Traditional Chinese Medicine on the treatment of male infertility is very effective, which is confirmed by ancient physicians.With the pharmacological mechanism of in-depth study of Chinese medicine, herbal active ingredients-the monomers in the treatment of male infertility has gradually begun, particularly in animal experiments.Based on modern chinese medicine theory, we choose lycium barbarum polysaccharides to increase lower T, LH, FSH levels, icariin to stimulate T secretion, which the former produces a multi-level regulatory role on the hypothalamus-pituitary-gonadal axis and the latter plays a major role in the testicles with little effect for the hypothalamus and pituitary,considering the compatibility between the two applications;supplemented with resveratrol of blood circulation drugs resist oxidation, scaveng oxygen free radicals, improve their hygiene microscopic environment and promote spermatogenesis and improve sperm dynamic effect; at the same time, three kinds of traditional chinese medicine which have plant estrogen-like effect on estrogen levels have a two-way adjustment, fully adjust the E2 level during spermatogenesis in the testis. Therefore, guiding the spirit of modern chinese medicine theory and the theory of reproductive endocrine hormones on the regulation of sperm production, using three kinds of traditional chinese medicine monomer, and L-toxic for additonal to explore the effect of the chinese herbal monomer and its' compounded on spermatogenic disturbance in mice induced by 60Co-γ.Methods:105 male Kunming mice were randomly divided into seven groups. Group A(n=15) were normally raised and group B, C, D, E, F, and G (15 each) were irradiated by 60Co-γ6 Gy and after 7 days by 60Co-γ4 Gy. A week, group C, D, E, F and G were given respectively resveratrol suspension liquid, resveratrol+LBP suspension liquid, resveratrol+icariin suspension liquid, resveratrol+LBP+icariin suspension liquid and resveratrol+LBP+icariin+L-carnitine compound suspension liquid with 80 mg/(kg·d) for 60 days. the general condition physical sign and body weight of mice were recorded,and 24 hours after finishing gavage, the weight of testes were measured and the testicular organ index were calculated; the levels of FSH, LH, T and E2 determined by enzyme-linked immunosorbent assay and the ratio of T/E2 were calculate; HE staining pathology of testicular is observed,also the c-kit expression on the spermatogonial cell surface by immunohistochemical SABC assay.Results:After the 60Co-γirradiation, the mouse showed the spirit of many apathetic, dull hair messy, hating cold and curling up,more seriously gone to death,also the testicular index fallen(P<0.05).The general situation and testicular index recovered to the normal at the end of perfusion,especially for group E,F and G.The levels of FSH,LH and T were obviously improved by LBP(P<0.05). The levels of T and the. weight of testes were increased by Icariin directly(P<0.05). The levels of T/E2 were increased in Group E, F, G(P<0.05); group F was the best and made the reproductive endocrine disorder close to normal,promated the recovery of spermatogenesis,and the surface markers c-kit of the spermatogonia restarted to express;but the group F and G showed no significant difference(P>0.05).Conclusion:the Chinese medicine compounded is effective for spermatogenic disturbance in mice induced by 60Co-y, and group resveratrol+LBP+icariin compound suspension liquid is the best; L-carnitine has not played the promoting role of spermatogenic.
