Objective: In-depth exploration of the “hepatorenal homologous” theory,Observing the effect of aerobic exercise as a prescription for “liver-regulating” and supplementing Kidney-nourishing Chinese herbs Drynaria Fortunei on osteoporosis in ovariectomized rats,to provide theoretical support and experimental basis for the prevention and treatment of clinical osteoporosis.Materials and methods: 45 female SD rats of 180 days of age as subjects in the experiment from July to October 2017,the body weight(200-209)g,average(204+2.5)g,was selected,adapted feeding for a week before the experiment begin.Then randomly and evenly divide the rats into 5 groups,Group ?: sham operation group(9 rats);Group ?: model group(9 rats);Group ?: aerobic exercise group(9 rats);Group ?: Drynaria Fortunei group(9 rats);Group ?: Drynaria Fortunei combined with aerobic exercise group(9 rats).In the sham operated group,only part of the adipose tissue adjacent to the ovary was excised(isosomal size),and the other four groups were treated by bilateral ovariectomy(OVX)to establish an animal model of ovariectomized osteoporosis in rats.One week after ovariectomy,rats were given Chinese medicine and/or aerobic exercise intervention according to group conditions.The traditional Chinese medicine Drynaria Fortunei free decoction added water to make 100%(1g/ml)concentration,Group ? and Group ? were fed 4ml of water per day(the same source of water uaysed for dispensing),rats in group ? and group ? were given intragastric herbal decoction solution(1g/ml)4ml daily,rats of group ? and group ? were subjected to aerobic exercise on the experimental animal “water wheel fatigue tester” at a speed of 12 m/min and a duration of 20 min.Rats in Group ? and Group ? performed aerobic exercise on the “Water Wheel Fatigue Meter” of experimental animals.The initial speed in the first week was 12m/min and the duration was 20 minutes.The intensity was increased by 1m/min every other day and the duration was increased 10 min.From the second week,the initial speed is 14m/min and the duration is 40 min.Each subsequent exercise maintains this strength.Exercise for 5 days a week,2 times a day,morning and evening.On rest days,rats in each group took a free diet.After continuous intervention for 12 weeks,the rats were obtained and anaesthetized by intraperitoneal injection of 0.3% chloral hydrate at a concentration of 0.3% per 100 g of body weight.Blood was collected from the abdominal aorta and the left femur of the rat was taken.The levels of alkaline phosphatase(ALP)and tartrate-resistant acid phosphatase(TRACP)in serum samples of each group were determined by enzyme-linked immunosorbent assay(ELISA).Serum calcium(Ca)and phosphorus(P)concentrations were measured using an automatic biochemical analyzer.Rat femur bone mineral density(BMD)was measured using a dual-energy X-ray absorptiometer(DEXA)small animal model.Based on the analysis of the“hepatorenal homologous” theory,aerobic exercise is used as a“liver-regulating”prescription combined with Bushen Chinese herbal supplement to prevent and treat osteoporosis in ovariectomized rats.Results: 1.After 12 weeks of various interventions,rats in Group ? and ? responded responsive and quick action.Their body hair was bright and smooth.Rats in Group ? and Group ? had a slower reaction rate than Group ? and ?,and the hair was lusterless and occasionally rough.Rats in Group ? were generally insensitive to reaction and had dry,dull hair.2.Comparison of BMD in each group: compared with Group ?,Group ?,?,? was significantly reduced,P<0.01,this result has extremely significant statistical differences.No statistical difference between Group ? and ?,P>0.05.Compared with Group ?,Group ?,?,? were all elevated,P<0.01,which was a very significant statistical difference.The results of Group ? and ? are basically the same,P>0.05,no statistical difference.Group ? was higher than Group ?,P<0.01,with extremely significant statistical difference.Group ? was lower than Group ?,P<0.01,with a very significant statistical difference.3.Comparison of serum ALP levels in each group: compared with Group ?,Group ? significantly decreased,P<0.01,there was a very significant statistical difference;Group ? decreased slightly,p<0.05,there was a statistical difference;Group ? and Group ? were basically the same,p>0.05,no statistics Differences in learning;Group ? increased,p < 0.01,the difference was statistically significant;compared with Group ?,Group ?,? and ? were significantly higher,p <0.01,there was a very significant statistical difference;Group ? compared with group ?,p<0.05,the difference was statistically significant.Group ? significantly increased,P<0.01,there was a very significant statistical difference;Group ?,?,? significantly reduced,p<0.05,there was a very significant statistical difference;Compared with Group ?,the Group ?,? and ? significantly decreased,P <0.01,there was a very significant statistical difference;Compared with Group ?,Group ?,? was significantly lower,P <0.01,the difference was statistically significant;Group ? was higher than Group ?,P <0.01,there was a very significant statistical difference.5.Comparison of serum Ca levels in each group: compared with Group ?,Group ?,?,?,and ? were significantly lower(P<0.01),and there was a statistically significant difference.Compared with Group ?,the serum Ca levels in Group ?,?,and ? were significantly higher,P<0.01,there was a significant statistical difference;Group ? compared with Group ?,P<0.05,there was a significant statistical difference;Compared with Group ?,serum Ca levels of Group ?,? decreased,P<0.05,There are significant statistical differences.6.Serum P levels were similar between groups,P>0.05,no statistical difference.Conclusion: 1.The increase of mental state of ovariectomized rats after intervention of aerobic exercise may be related to the exertion of aerobic exercise to promote the physiological characteristics of “liver governing regulating.” 2.Chinese Herbs Drynaria Fortunei combined with aerobic exercise has a good preventive effect on osteoporosis in ovariectomized rats and helps to improve potential mental state of rats,improve bone mineral density.Bone formation is promoted by increasing ALP activity,bone resorption is inhibited by decreasing TRACP activity,and bone mineralization rate is increased by increasing serum Ca content.3.Aerobic exercise can be used as a special prescription to play a “liver-regulating” role in the application of “hepatorenal homologous” theory.
