Based On The MTOR Signal Pathway To Explore The Mechanism Of Buzhengxugu Pills On PMOP And Osteoporotic Fractures

Purpose:To Objective: to establish PMOP and osteoporotic fracture syndrome animal model,drug intervention,the bone density,bone metabolism are closely related to the expression of peripheral blood serum CTX,the content of P1 NP and mTOR1 signaling pathways downstream of autophagy related protein p70S6 K,4EBP1 as observation indexes of pill on postmenopausal osteoporosis the density and the expression of p70S6 K and 4EBP1 effect.To explore the mechanism of the treatment of PMOP and osteoporotic fracture by Bu Zheng continued bone pill.Material and method:100 healthy female rats were divided into 10 groups by random number table method,10 rats in each group: normal control group,model group,sham operation group,Chinese medicine group of high,medium and low dose,the western medicine group(ALN)and Chinese medicine control group,pathway inhibitor group(rapamycin).The normal control group,sham operation group and model group were filled with pure water.Weigh it once a week and adjust it to the dose.After 6 weeks of administration,BMD,P1 NP,beta-CTX and so on were measured.The results are mean standard deviation of that using single factor analysis of variance,the comparison between groups and test.Results:1.BuZheng XuGu pill has some preventive effect on PMOP.2.BuZheng XuGu pill can reduce the content of β-CTX in the rat serum,which indicates that Bu Zheng continued bone pill can inhibit bone absorption.And have no obvious effect on P1 NP,3.PMOP is associated with the increase of the expression of p70S6 K and 4EBP1 in the mTOR1 signaling pathway.BuZheng XuGu pill has a certain inhibitory effect on the expression of p70S6 K and 4EBP1 in the mTOR1 signaling pathway.4.BuZheng XuGu pill can promote healing of osteoporotic fracture and improve the density of callus.5.The mechanism of the treatment of PMOP and osteoporotic fracture by BuZheng XuGu pill may be related to the expression of p70S6 K and 4EBP1 in the inhibition of autophagy related mTOR1 signaling pathway.Conclusion:BuZheng XuGu pill has some preventive and therapeutic effects on PMOP and osteoporotic fracture,and it can also inhibit bone resorption by regulating autophagy related mTOR1 signaling pathway p70S6 K and 4EBP1 expression.The specific mechanism is worthy of further study.This article only carries out the experimental part of the animal,and the cell experiment will be carried out in the future.
