Purpose: Elsholizia is originated from dried aerial parts of Mosla chinensis Maxim or Mosla chinensis‘Jiangxiangru' in Family Labiatae,the earliest recorded is from the "famous doctor don't record",as medium-grade drug,also was known as soft,pungent,tepor,it can sweat,dampness and moderate,inducing diuresis to alleviate edema.All the chinese Pharmacopoeia collected.The traditional processing removed residual roots and impurities,drying,softening,cutting,drying,complicated,easily lead to the change of the composition,and loss of effective components.Therefore,this topic will relate origin processing to processing and produing,clean in production area,freshly cut and dry to improve its quality.We do the research of equalization and equivalence of integration processing of origin and processing of traditional cutting technology to verify the feasibility of integration processing of origin technology,based on the study of rats with spleen and stomach dampness effect,provide the basis for the modern pharmacology of Elsholtzia in the wet and traditional effect,in order to provide reference for clinical rational use of drugs.Material and methodMaterial: Elsholtzia was collected in Jiangxi Xinyu and Hunan Changsha in September 2016.The herb was identified by Prof.Haibo Yin in School of Pharmaceutical Sciences,Liaoning University of Traditional Chinese Medicine,dalian,China.Method1.The traditional cutting processing system with the origin of comparative study of Mosla chinensis MaximBy L9(34)orthogonal test,taking the softening time,the length of cutting section and the drying temperature as the factors of the traditional cutting process,the cutting length,drying temperature and drying time were taken as the investigation factors of the integrated producting and processing technology,with carvacrol,thymol,volatile oil content as the evaluation index,to optimize the traditional cutting processing and the origin of the integration process,to compare the two selected process,provide the feasible basis for the implementation of processing of origin integration technology.2.Pilot study on integrated processing of Mosla chinese MaximOn the laboratory on the basis of the data,according to the selected cutting and drying technology in Jiangxi 100 kg/batches of Elsholtzia,pilot study was conducted in a total of 3 batches,namely while fresh cut 1 cm,36 h 50?~60?drying,enlarge research,so as to explore the feasibility of the implementation of the habitat processing technology integration.3.Comparing the content of phenols and flavonoids of the integrated producting and processing with traditional processing of Mosla chinese MaximThe kaempferol,carvacrol,thymol and other phenolic compounds and luteolin,apigenin,baicalein-7-ether and other flavonoids are indexed to compare the integrated producting and processing with traditional processing to provide a feasible basis for the implementation of processing of origin integration technology on the chemical level.4.GC-MS analyses the effect of volatile oil of Mosla Herba which was prepared by integration processing technology of originThe method of GC-MS was used.Chromatographic column: HP-5MS quartz capillary column(30m×0.25mm×0.25?m).Column temperature: initial temperature 60?,remaining 2 min,heating to 190 ? at 5?/min,remaining 6 min,heating to 250? at 5?/min,remaining 5 min;The carrier gas is helium;50:1 split injection;The inlet temperature is 250?;The column flow: 110?/min;Comparing the results of GC-MS and laying a foundation for the implementation of processing of origin integration technology.5.A comparative study on the effect of LPS on hot ratsCompared with the model group,the body weight of the rats increased,the temperature of the anus decreased,and the content of PGE2?TNF-??CAMP?IL-1??MPO decreased,and whether the volatile oil or water decoction,are IPTO has a significant effect,the difference was statistically significant(P <0.05),and showed a significant dose response relationship;6.A comparative study on the effect of i lung-Yang deficiency ratsLung-Yang deficiency rats were established with three-factor combination,swimming in common and ice water and drinking ice water,Continuously was treated for 30 days.The general signs,Anal temperature and autonomic activity of the rats were observed.The pathological morphology of lung tissue was observed,and the positive expression of TNF-? was observed by immunohistochemical method,and the hematological indexes were determined.And ELISA method was used to detect serum nitric oxide(NO),immunoglobulin G(IGG),malondialdehyde(MDA),thromboxane B2(TXB2)and interleukin-8(IL-8)level,calculating the heart liver spleen lung,kidney and other organ coefficient to analyze the therapeutic effect of Mosla chinese Maxim on lung-Yang deficiency rats.7.A comparative study on the effect of lung-Yin deficiency ratsLung-Yin deficiency rats were established with Thyroid Tablets and smoke,were given by gavage for 30 days.The general signs,Anal temperature and autonomic activity of the rats were observed.The pathological morphology of lung tissue was observed,and the positive expression of TNF-? was observed by immunohistochemical method,and the hematological indexes were determined.The abdominal aorta was taken for blood to detect urea(BUN),albumin(ALB),total protein(TP)and cholesterol(CHO).And ELISA method was used to detect interleukin-1?interleukin-1??cyclic adenosine(CAMP)and cyclophosphate(CGMP)level,calculating the heart liver spleen lung,kidney and other organ coefficient to analyze the effect of Mosla chinese Maxim on lung-Yin deficiency rats.8.A comparative study on the effect of ears swollen mice and RAW264.7 inflammatory cellsKM male mice were randomly divided into 8 rats in each group,which were given medicine by gavage for 7 days,ear swelling model was duplicated and swelling degree and swelling inhibition rate were calculated.With different concentrations of Moslae chinese Maxim volatile(integration processing of origin and processing of traditional cutting),Decoction(integration processing of origin and processing of traditional cutting),volatile oil and water extract mixture(crude drug equivalent)(integration processing of origin and processing of traditional cutting)on normal and LPS activated macrophages RAW264.7.To find out the best concentration,and to study the survival rate?phagocytic rate and NO index of every samples for LPS stimulated RAW264.7 cells.Providing reference for clinical rational use of drugs by researching the anti-inflammatory effect of Elsholtzia.9.The effect of Elsholtzia on the rat model of dampness obstructing spleen stomach syndromeComparing with the model group,rats body weight,food intake,water intake,urine volume,spleen and thymus organ coefficient,gastric emptying and small intestinal propulsion rate increased in elsholizia volatile oil and decoction,stool wet weight and intestinal water content decreased,the serum Na+-K+-ATP enzyme,the activity of AMS increased,the content of GAS,Ig G increased,the content of MTL,IL-6,AQP2 reduced,the difference was statistically significant(P<0.05);the rate of small intestine and intestinal function of water and MTL water decoction on rats is slightly better than that of volatile oil,other indexes are volatile oil on rats significantly.Results:1.The traditional cutting processing system with the origin of comparative study of Mosla chinensis MaximThe integration of origin process: cutting to 1 indicator of medicinal components in Moslae Herba,the contents of carvacrol and thymol were higher in integration processing technology of origin of Moslae Herba.There are 7 kinds of non common components in Hunan and Jiangxi Moslae Herba,except 2-borneol is unique components in Hunan,the other 6 chemical components were detected only in Jiangxi Moslae Herba.5.A comparative study on the blood cell and in the lung tissues of rats with TNF-alpha positive area and mean optical density,the difference was statistically significant(P <0.01 or P <0.05),and whether the volatile oil or water decoction,are IPTO has a significant effect.7.A comparative study on the effect of lung-Yin deficiency ratsElsholtzia improves the general signs of autonomic activity effect is not obvious for lung-yin deficiency rats,has little effect on the body weight,total protein and albumin and heart,liver and spleen,lung,kidney coefficient,has little influence on serum IL-1,IL-1,CAMP,CGMP and other indicators of beta and rectal temperature.And reduces the total expression of TNF-alpha and the average optical density of the trend of little effect on lung-Yin deficiency rats is not obvious.8.A comparative study on the effect of ears swollen mice and RAW264.7 inflammatory cellsIn vivo ear swelling model,compared with the model group,each group has a sample of Elsholtzia treatment of ear swelling effect,and the effect of group integration processing of Elsholtzia origin decoction best in RAW264.7 cell model of inflammation induced by LPS in vitro in each sample group were Elsholtzia with enhanced cellular immune function,and to the integrated processing of Elsholtzia origin decoction group obvious effect.9.The processing of Elsholtzia on the rat model of dampness obstructing traditional cutting method.The feasibility of the integrated processing technology was proved by the equal quantity of chemical composition.The integration of processing Elsholtzia was better than the traditional cutting method on LPS induced rats,lung Yang deficiency and lung yin deficiency rats,ear swelling mice and RAW264.7 inflammatory cells.Based on the five experiments,it is verified that the quality of traditional cutting Elsholtzia is better after the integration of processing.Through the study of integration of processing Elsholtzia,it is proved that the integration of chemical composition and pharmacological action of the Elsholtzia can reduce the loss of the ingredients and improve the curative effect of the pieces,and it has the feasibility to implement.The effect of Elsholtzia on dampness and spleen stomach syndrome was studied,and it was found that it had good humidification and medium effect.
