PurPose:To observe Qingzhi Tongmai Granules on blood lipid levels and expression levels of microRNA33(miR-33),ATP binding cassette transporter A1(ABCA1)and ATP binding cassette transporter G1(ABCG1)in experimental atherosclerosis model rats The impact of the further observation of the links between the indicators,and the optimal ratio of prevention and treatment of atherosclerosis drugs.Material and method:1.144 SPF rats were selected as the research object of this experiment,and 144 rats were randomly grouped by random number table method,12 in each group,namely normal group,model group and Chinese medicine group 12 groups.12 groups.After one week of adaptive feeding in all experimental rats,except for the normal group,the other groups were modeled.The specific method was to inject vitamin D3+ compound high-fat diet,and the normal group was injected with the same amount of normal saline and fed with conventional feed.2.After 9 weeks of the experiment,samples were taken from the rats in each group,and the contents of serum cholesterol(TC),triglyceride(TG),low-density lipoprotein(LDL)and high-density lipoprotein(HDL)were detected.The expression levels of corresponding genes of mir-33,ABCA1 and ABCG1 in liver were detected by PCR,and the expression levels of ABCA1 and ABCG1 were detected by Western-blot.3.The results of Q-PCR showed that the genic expression level of miR-33 in the liver was increased in the model group and the Qingzhi Tongmai granule group(P<0.05).ABCA1,ABCG1 genic expression levels in the liver were reduced(P<0.05).Compared with the model group,the genic expression level of miR-33 in the liver was decreased(P<0.05),and the genic expression level of ABCA1 and ABCG1 in the liver was increased(P<0.05).Results:1.The results of blood lipid test in each group showed that compared with the normal group,the contents of TC,TG and LDL in the model group were significantly increased(P<0.01),and the HDL-C content was significantly decreased(P<0.05).Qingzhi Tongmai Granules canreduce the TG content in serum(P<0.01).In the detection of TC,we found that the contents of groups 3,7,8,9,and 12 were significantly lower(P<0.05).The HDL-C content of 5,6,8,11 and 12 increased significantly(P<0.05).After the test,the LDL-C content was also significantly reduced in groups 3,7,8,10,and12.The levels of TC,TG and LDL-C in the group were not significantly decreased,and the HDL-C content was slightly increased,but it was not statistically significant(P>0.05).2.The results of Western-blot in each group showed that compared with the normal group,the protein expression levels of ABCA1 and ABCG1 in the liver of the model group were decreased(P<0.05).Compared with the model group,the expression levels of ABCA1 and ABCG1 in the liver were increased(P<0.05).3.Q-PCR results showed that compared with normal group,model group,fat eliminatin g pulse particle group miR-33 in the liver gene expression levels(P<0.05),ABCA1,ABCG1 gene expression level in the liver significantly decreased(P<0.01).Compared with the model group,the gene expression level of mir-33 in the liver of qingzhi ton gmai granules group was decreased(P<0.05),the gene expression level of ABCA1 in the liver was significantly increased(P<0.01),and the gene expression level of ABCG1 in the liver was increased(P<0.05).Conclusion:1.It can be known from blood lipid test that group 8(Optimal dose group 1)and group 12(Optimal dose group 2)are superior to other traditional Chinese medicine groups in lipid-lowering effect.2.The use of Qingzhi Tongmai Granule can inhibit the expression of miR-33 gene in the liver tissue of model rats,and can promote the expression of ABCA1,ABCG1 gene and protein.3.The reverse transport of cholesterol may be achieved by using Qingzhi Tongmai Granule to inhibit the expression of miR-33 gene in liver tissue of model rats and increase the gene and protein expression levels of ABCA1 and ABCG1.4.The mechanism of Qingzhi Tongmai Granules in preventing and treating AS may be through inhibiting the expression of miR-33 gene in liver tissue,increasing the expression levels of ABCA1,ABCG1 gene and protein,thereby accelerating the rate of cholesteroloutflow to peripheral tissues and promoting the reverse transport of cholesterol.
