| Objective:To study the effect of acupuncture-drug binding on the expression of GABAa receptors and synaptic plasticity-related proteins GAT-1、PSD95 cortical area of stroke limb spasm rats and to explore the mechanism of acupuncture-drug binding in the treatment of stroke limb spasm.Methods: The 129 male SD rats were randomly divided into blank group,9sham operation group and 111 model reserve group.The modified Zea longa line internal capsule injection NMDA method was used to make the rat model of stroke limb spasm.The 45 rats were randomly divided into five groups: model group,electroacupuncture group(electroacupuncture Yanglingquan,qu Chi),traditional Chinese medicine group(filling with paeoniflorin liquorice decoction),western medicine group(filling with baclofen solution)and combination of acupuncture and medicine group(electroacupuncture Yanglingquan,qu Chi adding paeoniflorin liquorice decoction).Zea longa nerve damage score was used to evaluate the nerve function,the modified Ashworth muscle tension score was used to evaluate the muscle tension,the changes of synaptic morphology and structure were observed by electron microscope,the GABAa receptor,GAT-1、PSD95m RNA expression of rat cerebral cortex were detected by Rt-PCR method,and the GABAa receptor and GAT-1、PSD95 protein expression of rat cerebral cortex were detected by the method.Results: 1.Zea longa nerve damage score,modified Ashworth muscle tension score: before treatment,compared with the blank group,there was no difference(P>0.05)in the sham operation group.compared with the blank group and sham operation group,the score of the model group was increased,which was statistically significant(P<0.01),suggesting that the stroke limb spasm model was successfully prepared;after treatment,the scores of each treatment group and the model group were all decreased,the difference was statistically significant(P<0.05,P<0.01),and there was no difference between the groups(P>0.05).The results showed that the combination of electroacupuncture,traditional Chinese medicine,western medicineand acupuncture could improve the nerve function and muscle tension of stroke limb spasm rats.2.Changes of synaptic morphology and structure under electron microscope: the number of synapses in the blank group was more,the synaptic vesicles were clear and clear,the anterior and posterior membranes were intact and the synaptic space was clear.Compared with electroacupuncture group,the Chinese medicine group has little difference,the acupuncture and drug combination group and the western medicine group have dense synapses,the number of synaptic vesicles is increased,the dense matter is thickened,and the gap is reduced;compared with the western medicine group,the acupuncture and drug combination group has little difference.3.The expression of GABAa receptor and GAT-1、PSD95m RNA in rat cortical area: compared with blank group and sham operation group,the expression of GABAa receptor and PSD95 m RNA in model group decreased significantly,and the expression of GAT-1m RNA increased significantly,and the difference was statistically significant(P<0.01);The expression of GABAa receptors and PSD95 m RNA increased and the expression of GAT-1m RNA decreased(P<0.05);The expression of GABAa receptors and PSD95 m RNA in western medicine group and acupuncture-drug binding group increased and GAT-1m RNA decreased compared with electroacupuncture group(P<0.05);The expression of GABAa receptor and PSD95 m RNA in western medicine group and acupuncture-drug binding group was higher and the expression of GAT-1m RNA decreased(P<0.05).4.The expression of GABAa receptor and GAT-1、PSD95 protein in the cortical area of rats: compared with the blank group and the sham operation group,the expression of GABAa receptor and PSD95 protein in the model group was significantly decreased and the difference was statistically significant(P<0.01);GAT-1 decreased GABAa receptor and PSD9 protein expression in treatment groups compared with model group(P<0.05);The expression of GABAa receptors and PSD9 proteins in western medicine group and acupuncture-drug binding group decreased GAT-1 compared with electroacupuncture group(P<0.05);The expression of proteinin western medicine group and acupuncture-drug binding group GABAa increased PSD95 and decreased compared with traditional Chinese medicine group(P<0.05).Conclusion: 1.Combination of acupuncture and medicine can improve nerve function and muscle tension in stroke limb spasm rats;2.acupuncture and drug binding can increase the expression of GABAa receptors and PSD95 in the cortical area of stroke limb spasm rats,and decrease the expression of GAT-1.it is shown that the mechanism of acupuncture and drug binding to alleviate stroke limb spasm may be through the regulation of rat cortical area GABAa receptors,synaptic plasticity related proteins. |
