| Objective: This study was to observe the effect of Yiqi Dingxuan drink on the neural function of rats with cerebral Ischemia,and to explore whether BDNF / Trk B pathway plays a neuroprotective role.Methods:120 clean male healthy adult rats were randomly divided into Sham Operation Group,Model Group,butylphthalide control group and Yiqi Dingxuan drink treatment group.The MODEL OF CEREBRAL ISCHEMIA was made by modified middle cerebral artery occlusion(MCAO).The sham-operation group was not plugged,and the rats were given routine nursing after operation.The Control Group and Treatment Group were given butylphthalide and Yiqi Dingxuan drink respectively,the rats in the sham-operation Group and the model group were given intragastric distilled water once a day for 1,3,7 and 14 days to evaluate the neurological function of the rats,and the hippocampal tissue sections were taken from the rats,and the histopathology of the hippocampus was observed by HE staining,the expression of BDNF and Trk B in hippocampus was detected by immunohistochemistry.Results:1.Compared with the model group,the scores of neurological deficit in Yiqi Dingxuan drink group and butylphthalide group decreased on the 3rd,7th and 14 th days after Operation(p<0.05),but there was no significant difference betweeni Yiqi Dingxuan drink group and butylphthalide group at the same time point(p>0.05).2.Compared with the model group,HE staining showed that the nerve cells in Yiqi Dingxuan decoction group and butylphthalide group were slightly disordered,the nucleus dissolution was reduced,the cell structure was intact and the number of cells increased.3.Results: compared with the model group on the 3rd,7th and 14 th days,the expression of BDNF and Trk B positive cells in the modified yiqiding group,the Yiqi Dingxuan Yin Group and the butylphthalide group increased(p<0.05).However,there was no significant difference between Jiawei Yiqi Dingxuan drink group and butylphthalide group(p>0.05).Conclusion: The Yiqi Dingxuan drink can improve the neurologic deficit of cerebral ischemia rats and reduce the injury of neuron cells.The effect of Yiqi Dingxuan decoction is obvious after 3 days intervention,which indicates that Yiqi Dingxuan decoction has protective effect on hippocampal neurons of MCAO rats,the mechanism may be that BDNF / Trk B signaling pathway up-regulates the expression of BDNF and its receptor Trk B after cerebral ischemia injury. |
