Objective:The dried and mature seeds of Herpetospermum caudigerum Wall.a family of Cucurbitaceae plants,are commonly used Tibetan medicine and are collected by the Ministry of Health of the People’s Republic of China in the pharmaceutical standard Tibetan medicine(Volume 1).Boleng melon seeds taste bitter,nature cold,can clear the viscera heat,gallbladder heat;treatment of red bar into the viscera,liver,gallbladder fever(hepatitis,cholecystitis),indigestion.A large number of studies have shown that lignans are the main components of liver protection,and lignans have certain protective effects on liver fibrosis and liver injury。Alcoholic liver injury(ald)mainly refers to alcoholic fatty liver,alcoholic liver fibrosis,alcoholic hepatitis,and toxic liver damage caused by chronic heavy alcohol consumption.Modern medical treatment of ALD mainly includes abstinence from alcohol and symptomatic treatment,other treatments are less.In recent years,Chinese medicine has more research results on prevention and treatment of ALD in the experimental research of single traditional Chinese medicine extract and the application of clinical prescription.The literature shows that the model of acute alcoholic liver injury can be treated by the total number of melon seeds The protective effect of total lignans on liver tissue and liver function injury in acute alcoholic liver injury rats may be to decrease the expression of TGF-beta;protein and decrease the content of CTGF.Therefore,the mechanism of lignin in treating alcoholic liver injury in TGF-beta;Smads and CTGF pathways may provide a new idea for the treatment of alcoholic liver injury.Method:1.Lignans from seeds of Herpetospermum caudigerum Wall.protect against alcoholic liver injury via TGF-β/Smads pathway in rats:60 SD rats were randomly divided into six groups,except for the normal group,the model of alcohol-induced liver injury was established for 8 weeks,and the model was simult aneously modeled at 400,200,100 mg/kg in the high and low dose group of lignin,and the control group was given 50 mg/kg of silimarin once a day for 8 weeks.TGF-β,TGF-TGF-β receptor 1(TβR-I),TGF-β receptor 2(TβR-II),Smad2/p-smad2,smad3/p-smad3 protein expression;elisa detected the content of TGF-β and CTGF in liver tissues;immunohistochemical staining observed the distribution of TGF-β.Results: The expression of TGF-β,Smad2,p-Smad2 and p-Smad3 proteins in rats with lignans and thistle group were significantly decreased,but the expression of TβR-I,TβR-II and Smad3 proteins were not significantly changed.TGF-β,Smad2,P-Smad3 in rats with thistle group the expression of TβR-I,TβR-II,p-Smad2 and Smad3 proteins all decreased significantly compared with the model group.2.Lignans from seeds of Herpetospermum caudigerum Wall.protect against alcoholic liver injury via TGF-β/CTGF pathway in rats:60 SD rats were randomly divided into six groups,except for the blank group,the model of alcohol-induced liver injury was established for 8 weeks,and the model was simultaneously modeled at 400,200,100 mg/kg in the high and low dose group of lignin,and the control group was given 50 mg/kg ofsilimarin once a day for 8 weeks.The contents of TGF-β,CTGF and liver homogenate SOD in rat serum were measured by Elisa.The results of HE and Masson collagen staining in rat liver tissue were observed.Observe the distribution of CTGF.Result:1.Lignans from seeds of Herpetospermum caudigerum Wall.protect against alcoholic liver injury via TGF-β/Smads pathway in rats:(1)compared with the normal group,he staining and sirius red staining found that the model group had collagen fiber formation,and hepatocyte necrosis,hepatic sinus expansion around the central vein,accompanied by lymphocyte punctate infiltration.(2)Compared with the model group,the liver tissue injury of the rats in the group was significantly reduced,and the concentration of SOD was significantly increased(P <0.05,P <0.01).the expression of TGF-β,smad2,p-smad2 and p-smad3 proteins in rat liver tissues decreased significantly compared with the model group(P<0.05,P<0.01).the expression of TβR-Ⅰ、TβR-Ⅱ and Smad3 proteins in rat liver tissues of thistle group did not change significantly.2.Lignans from seeds of Herpetospermum caudigerum Wall.protect against alcoholic liver injury via TGF-β/CTGF pathway in rats:(1)Compared with the blank group,the liver of the model group was stained with HE and Masson three-color collagen,the model group had collagen fiber formation,the local manifold area showed biliary bile duct hyperplasia,and a small amount of lymphocyte infiltration were seen in the model group.(2)Compared with the model group,the liver coefficient was significantly decreased(P <0.05,P <0.01),the contents of TGF-beta and CTGF were also significantly decreased(P <0.05,P <0.01),and the content of liver plasma SOD was significantly increased(P <0.05,P <0.01),the degree of liver injury was reduced,and the formation of collagen fibers was reduced.Conclusion:In summary,the above in vivo experiments have shown that Pomegranite has obvious protective effect on liver tissue of alcoholic liver injury rats,has obvious repair effect on damaged liver tissue,and has obvious inhibitory effect on the formation of collagen fiber.The expression of TGF-beta,Smad2 and Smad3 in rats was significantly different by the treatment of ginseng resin in rats.The main reason was that TGF-\beta,Smad2 and Smad3 were significantly decreased in rats.whereas reduced CTGF expression mediated by TGF-beta,mediating the positive effect of TGF-beta and slowing the occurrence of liver injury towards hepatic fibrosis.In rats with liver injury,the inhibition effect of lignans increased with the increase of the dose of lignans.
