Purpose: The heart failure model was established by ligation of the left anterior descending coronary artery(LAD)in rats.The pharmacodynamics of the components of Qishen Granule,Wenyang Yiqi,Huoxue and Detoxification were evaluated and the heart failure myocardial infarction was studied.Research on the regulation mechanism of energy metabolism.Method: 1.Male SD rats were treated with LAD ligation for 1 week.The patients were divided into sham operation group(only threaded without ligation),model group(LAD ligation,gavage water),and phlegm particle group(LAD ligation).,sputum sputum granules),Wenyang group(LAD ligation,irrigation Wenyang Yiqi Chinese medicine),Huoxue group(LAD ligation,irrigation and blood circulation Chinese medicine)detoxification group(LAD ligation,irrigation and detoxification Chinese medicine),fosinopril group(LAD ligation,administer Fosinopril),take the medicine once a day,feed after 28 days,through echocardiography,HE staining for ginseng granules,Wenyang Yiqi Chinese medicine,blood circulation Chinese medicine,detoxification Chinese medicine and fosinopril Pharmacodynamic research.2.The blood lipid indexes TC,TG,LDL and HDL were detected by sham operation group,model group,Cushen granule group,Wenyang Yiqi group,Huoxue group,detoxification group and fosinopril group.3.P-AMPK and PGC-1α were detected by Western Blot in the sham operation group,model group,Cushen Granule group,Wenyang Yiqi group,Huoxue group,Jiedu group and Fosinopril group.4.The lipid metabolism related indexes PPARα,CD36 and CPT-1a were detected by sham operation group,model group,Cushen granule group,Wenyang Yiqi group,Huoxue group,detoxification group and fosinopril group by Western Blot method.Result: 1.According to the results of small animal echocardiography,the EF and FS values of the model group were significantly lower than those of the sham operation group,which were 52.83% and 45.83%,respectively(both P<0.01).The treatment of ginseng granules for 28 days can be significantly improved.The EF value and FS value of heart failure rats were increased by 14.09%(P<0.01)and 7.75%(P<0.01)compared with the model group.Wenyang Yiqi group,Huoxue group,detoxification group and fosinopril group After treatment,similar effects were observed.The improvement of EF and FS in Wenyang Yiqi group was 11.97% and 6.96% higher than that in model group(both P<0.01).The improvement of EF value and FS value in Huoxue group were compared with the model.The group increased by 12.10% and 7.77%(both P<0.01);the improvement of EF and FS in the detoxification group was 11.34%(P<0.01)and 5.94%(P<0.05),respectively.The improvement of EF and FS in the benefit group was 14.37% and 7.95% higher than the model group(both P<0.01).The model group LVID;d and LVID;s were significantly higher than the sham operation group,indicating that the heart group of the model group was enlarged.After treatment with Qishen Granules,cardiac cavity enlargement was significantly inhibited,LVID;d and LVID;s decreased by 17.17%(P<0.01)and 16.01%(P<0.01),respectively.Wenyang Yiqi group,Huoxue group,detoxification The LVID;d and LVID;s were down-regulated in the group and the fosinopril group compared with the model group.The Wenyang Yiqi group was 11.40%(P<0.05)and 20.31 lower than the model group LVID;d and LVID;%(both P<0.01);LVID;d and LVID;s decreased by 11.73%(P<0.05)and 15.85%(P<0.01),respectively;the detoxification group was compared with the model group LVID;d and LVID;s decreased by 8.52% and 15.29%,respectively(P<0.01).There was no significant difference between the detoxification group and the model group LVID;d;the fosinopril group decreased by 1.72 compared with the model group LVID;d and LVID;s 8.02%,the two groups of data were not significantly different from the model group.2.The heart tissue sections were magnified 100 times by HE staining,and the effects of each group on myocardial tissue of heart failure after myocardial infarction were observed in the visual field.Among them,the myocardial cells in the sham operation group were arranged in order and tight;in the model group,the myocardial cells in the marginal zone of the myocardial infarction could be seen to be loose,and the interstitial cells were replaced by a large number of fibroblasts and in the interstitial space.There are many inflammatory cell infiltrations;although the ginseng granules are also filled with fibrous tissue in the interstitial space,compared with the model group,the infiltration of fibers in the interstitial space is limited,and the myocardial cells are densely arranged and larger.This allows cardiomyocytes to better maintain ventricular wall toughness.The effects of Wenyang Yiqi group,Huoxue group,Jiedu group and fosinopril group on cardiomyocytes of heart failure after myocardial infarction were similar to those of Fushen Granule group.3.The serum levels of TC,TG and LDL in the model group were significantly higher than those in the sham-operated group.After treatment with Qishen Granules for 28 days,the TC,TG and LDL were significantly reduced by 46.06%(P<0.01)and 60.98%(P< 0.01)and 72.31%(P < 0.01).The serum levels of TC,TG and LDL in the Wenyang Yiqi group were decreased by 43.95%(P<0.01),60.98%(P<0.01)and 50.55%(P<0.01),respectively.Compared with the model group,TC,TG and LDL decreased by 30.70%(P<0.01),29.55%(P<0.01)and 35.96%(P<0.01),respectively.Compared with the model group,TC,TG and LDL decreased by 34.16%(P<0.01),53.57%(P<0.01)and 42.63%(P<0.01).The TC,TG and LDL in the fosinopril group were 18.48%,45.23%(P<0.01)and 46.59%(P<0.01),respectively.The change of serum TC content in the fosinopril group compared with the model group.There were no significant differences in the model group.The serum HDL level in the model group was significantly lower than that in the sham operation group.The serum HDL level was 100.83% higher than that in the model group after treatment with Qishen Granule(P<0.01).The Wenyang group was 107.84% higher than the model group(P<0.01).The blood-activating group was 43.77% higher than the model group,and there was no significant difference in the content change;the detoxification group was 50.32% higher than the model group,and there was no significant difference in the content change;the fosinopril group was 73.34% higher than the model group(P< 0.01).4.The results of WB experiment showed that the P-AMPK of the model group was significantly decreased compared with the sham-operated group.Compared with the model group,the P-AMPK of the Qishen Granules increased by 113.29%.P < 0.01).The Wenyang group increased by 157.82% compared with the model group(P<0.01),the blood circulation group increased by 45.38%(P<0.01),and the detoxification group increased by 83.00% compared with the model group(P<0.01).The positive drug fosinopril group increased by 149.54% in the relative model group(P<0.01).Compared with the sham-operated rats,PGC-1α in the model group decreased significantly,and PGC-1α increased by 105.12%(P<0.01)after treatment with Qishen Granule.The Wenyang group and the detoxification group increased with respect to the model group.177.47%(P<0.01)and %122.82(P<0.01),the blood circulation group increased by 89.51% compared with the model group(P<0.01),and the fosinopril group increased by 117.43% compared with the model group(P<0.01).Compared with the sham-operated rats,the PPARα of the model group decreased significantly.Compared with the model group,the PPARα increased by 58.50%(P<0.01),and the PPARα of the Wenyang Yiqi group increased by 104.06%(P<0.01).The PPARα increased by 72.82%(P<0.01),the PPARα of the detoxification group increased by 163.66%(P<0.01),and the PPARα of the fosinopril group increased by 31.46%(P<0.01).Compared with the sham-operated rats,the CPT-1a content in the model group was significantly lower than that in the model group.The CPT-1a in the ginseng granule group increased by 122.86%(P<0.01),which indicated that the fatty acid metabolism decreased in the model group.Compared with the model group,the CPT-1a in the Wenyang group increased by 165.77%(P<0.01),and the Huoxue group increased by 108.29%(P<0.01).CPT-1a in the detoxification group increased by 153.55%(P<0.01),and the fosinopril group decreased by 20.81%.Compared with the sham-operated rats,the CD36 of the model group decreased significantly.Compared with the model group,the CD36 of the ginseng granules increased by 24.48%(P<0.01).Compared with the model group,CD36 in the Wenyang group increased by 29.06%(P<0.01),CD36 in the blood group increased by 24.42%(P<0.01),and CD36 in the detoxification group increased by 11.16%.There was no significant difference between the detoxification group and the model group.The CD36 of the Fosinopril group increased by 16.99%(P<0.05).Conclusion 1.QSG and its decomposed prescriptions Wenyang Yiqi group,Huoxue group and Jiedu group can effectively relieve the heart function of heart failure rats after heart failure.The FS value of FS is higher than that of the model group.For LVID;d and LVID;Both have improved to varying degrees.2.QSGcan effectively reduce the levels of TC,TG and LDL in the blood of rats with heart failure after myocardial infarction,and improve the HDL content in the blood,improve the heart metabolism and lipid metabolism disorder after myocardial infarction,and its decomposed components reduce blood TC.TG,LDL,and HDL all exert different degrees of effect.Among them,Wenyang Yiqi group is better in reducing blood TC,TG,LDL and improving HDL than other decomposed groups.3.QSG can effectively improve the energy metabolism of myocardial energy in rats with heart failure after myocardial infarction.The Wenyang Yiqi group in the decomposed group is more effective in improving energy metabolism,and can significantly improve the energy metabolism related indicators P-AMPK,PGC.-1α,which plays a major regulatory role in the whole party.4.QSG can effectively improve the level of myocardial lipid metabolism in rats with heart failure after myocardial infarction.The decomposed group can enhance the expression of lipid metabolism related proteins PPARα,CD36 and CPT-1a,and the Wenyang Yiqi group is more Prominent,play a major therapeutic role in the treatment of myocardial infarction after myocardial infarction,abnormal lipid metabolism,and elevated lipid metabolism. |
