Acupoint application is a unique therapy of traditional external treatment of traditional Chinese medicine.It is a kind of non-invasive acupoint therapy for preventing and treating diseases,which is based on the meridian theory of traditional Chinese medicine,grinds Chinese herbal medicine into powder,mixes traditional Chinese medicine powder into paste with water,vinegar,wine,honey,vegetable oil,vaseline,medicine liquid and other media,and then directly paste it to acupoints or affected areas.The theory of traditional Chinese medicine holds meridians have the functions of connecting viscera,communicating inside and outside,running qi and blood,and nourishing the whole body.Applying drugs directly to acupoints can not only stimulate acupoints to stimulate the qi of meridians and collaterals,but also enable the drug effect to reach the viscera through the meridians to achieve the purpose of treating diseases.Clinically,it has been proved that acupoint application can effectively reduce the level of SUA in patients,thus preventing and treating HUA.Research members have done experimental research on acupuncture at Shu,Mu and Yuan points to interfere with HUA model rats.The results show that acupuncture at Shu,Mu and Yuan points can reduce the level of SUA in rats in different degrees.However,considering the poor medical compliance of acupuncture treatment for patients clinically,on the basis of previous studies,the advantages of external treatment of traditional Chinese medicine are brought into play,and acupoint application is adopted to interfere with Shu and Mu points of liver,spleen and kidney in HUA model rats to evaluate its therapeutic effect and preliminarily explore its mechanism.Objective:HUA is a metabolic disease caused by metabolic disorder of purine substance.Uric acid is the final product of purine substance metabolism.Increased synthesis or insufficient excretion can lead to HUA.In this study,we observed the effects of Shu and Mu points applied to liver,spleen and kidney on SUA,ALT,AST,XO,ADA,GD,HGPRT activity level changes and the general situation of rats and liver tissue morphology and structure in HUA model rats by applying the two-sided points of liver,spleen and kidney,namely,Shu and Mu points of liver,spleen and kidney.To evaluate the effect of applying Shu and Mu points on liver,spleen and kidney to interfere with HUA model rats,and preliminarily explore the possible related mechanism of applying Shu and Mu points on liver,spleen and kidney to treat HUA,so as to provide certain theoretical basis for acupoint application to treat HUA,and also provide clinical guidance for acupoint application to treat HUA in selecting and matching points.Methods:Thirty-two clean male SD rats were randomly divided into blank group,model group,sham acupoint application group and acupoint application group,with 8 rats in each group.From the beginning of the experiment,the rats in the other three groups were given 2 g/(kg·d)of potassium oxazinate by intragastric administration of 10 mL/kg to establish HUA model rats,and the blank group was given the same volume of normal saline once a day for 10 days.Ten days later,blood was taken from the tail vein of all rats,and the level of SUA was measured by automatic biochemical analyzer.Compared with the blank group,the level of SUA in the other three groups increased significantly(P<0.01),indicating the successful establishment of HUA model.During the application treatment,all rats were given intragastric administration once a day instead of once every other day.The blank group was fed normally without any treatment.The model group was fed normally,only grasping and pressing properly.The rats in the sham acupoint application group and acupoint application group were treated with vaseline application and acupoint application medicine application on bilateral Ganshu and Qimen,Pishu and Zhangmen,Shenshu and Jingmen,respectively,each time for 6 hours,once a day,5 times as a course of treatment,a total of 3 courses of intervention,rest for 1 day after each course of treatment for 1 8 days.Samples were taken after the application treatment,the levels of SUA,ALT and AST in serum were measured by automatic biochemical analyzer,the activities of XO and ADA in liver tissue were detected by enzyme colorimetry,and the contents of GD and HGPRT in liver tissue were detected by enzyme-linked immunosorbent assay(ELISA).Results:(1)The level of SUA:on the 10th day of modeling,compared with the blank group,the level of SUA in model group,sham acupoint application group and acupoint application group increased significantly(P<0.01).After application treatment,compared with the blank group,the level of SUA in model group and sham acupoint application group increased(P<0.01 or P<0.05),and the level of SUA in acupoint application group decreased(P<0.01).(2)The levels of serum ALT and AST:after application treatment,compared with the other three groups,the level of serum ALT in the acupoint application group increased(P<0.01).The level of serum AST had no significant change(P>0.05).(3)XO and ADA activity in liver tissue:after application treatment,compared with the blank group,the level of XO activity in the model group and the sham acupoint application group increased(P<0.05),and the level of ADA activity had no significant change(P>0.05).Compared with the model group and the sham acupoint application group,the level of XO activity decreased and the ADA activity increased in the acupoint application group(P<0.05 or P<0.01).(4)GD and HGPRT activity in liver tissue:after application treatment,compared with the blank group,the level of GD activity in liver tissue in model group and sham acupoint application group increased(P<0.01 or P<0.05),and the level of HGPRT activity had no significant change(P>0.05).Compared with model group and sham acupoint application group,the activity of GD in liver tissue in model group and sham acupoint application group decreased(P<0.01),and the level of HGPRT activity had no significant change(P>0.05).(5)Observation of liver tissue sections:after application treatment,under light microscope,the hepatocytes of the four groups of rats were arranged orderly and connected with each other,radially arranged with the central vein as the axis,the structure of hepatic lobules was clear and complete,and the arrangement of hepatocyte cords was regular.The structure and morphology of hepatocytes were normal,and no obvious pathological changes were found in the four groups.Conclusion:(1)The application of Shu and Mu points on liver,spleen and kidney can significantly reduce the level of SUA and can effectively improve the diet,excretion function,mental state and activity response ability of HUA rats,and has a good preventive and therapeutic effect on HUA.(2)The application of Shu and Mu points on liver,spleen and kidney has a significant effect on reducing uric acid.The mechanism may be that uric acid is generated less by inhibiting the activities of XO and GD in the liver tissue,thus reducing the level of SUA.
