Objective:To make a comprehensive statistics of Professor Zhang Hongchun’s traditional Chinese medicine prescriptions for the treatment of bronchial asthma,systematically analyze the rules of drug use,explore the relationship between core drugs and high-frequency drugs in the prescriptions,and draw the core prescriptions and experience of Professor Zhang Hongchun for the treatment of bronchial asthma.To establish a network of core prescription drug components and targets,and to explore the mechanism of Zhang Hongchun’s core prescription in the treatment of bronchial asthma.Method:Study 1:Collect the medical records of Professor Zhang Hongchun’s treatment of bronchial asthma by useing Traditional Chinese Medicine Inheritance Support System(TCMISS),use the frequency statistics method to get the sexual taste and meridian frequency of prescription Chinese medicine;use the association rule analysis method to get the frequency of drug combination use and the association rules between drugs,and get the core prescription of Professor Zhang Hongchun’s treatment of bronchial asthma;use the entropy accumulation of complex system The new prescription is obtained by core combination analysis of class.Study 2:Query the core prescription drug ingredients and target points in the database,get the intersection with the disease target of bronchial asthma,and use Cytoscape 3.7.2 the software constructs the network diagram of traditional Chinese medicine compound target,and obtains the preliminary core drugs,compounds and targets through topological property analysis.Then,the core targets are introduced into string to construct PPI network,and more effective genes of concern are obtained.Then,the enrichment analysis and KEGG pathway analysis of effective genes are carried out by using clego and David,and the possible key of Professor Zhang Hongchun’s treatment of bronchial asthma is obtained Target and action pathway.Result:Study 1:59 cases and 252 prescriptions were included in this study.The results of four gas analysis of prescription drugs showed that the frequency of warm drugs was the highest,followed by cold drugs,and the five kinds of drugs were mainly pungent,bitter and sweet,with the same proportion;the main viscera of drugs were lung,spleen,stomach and liver;the top 6 drugs in the order of frequency from high to low were Liquoric Root、Perilla Leaf、Common Coltsfoot Flower、Baical Skullcap Root、Perilla Fruit and Ephedra Herb;the number of support degree was set to 150(i.e.support degree=60%),with the confidence set to 0.9,31 commonly used drug combinations were obtained.After the association rules containing Glycyrrhiza were eliminated,the three drug patterns with the highest frequency were:Perilla Fruit-Perilla Leaf、Perilla Leaf-Common Coltsfoot Flower、Perilla Leaf-Cicada Slough.Combined with the above drug dosage frequency analysis,the core prescriptions of Professor Zhang Hongchun in the treatment of bronchial asthma were obtained:burn Ephedra Herb 10g、Common Coltsfoot Flower 15g、Perilla Fruit 15g、Perilla Leaf 15g、Tatarian Aster Root 15g、Cicada Slough 8g、Baical Skullcap Root 15g、Great Burdock Achene 10g、Indian Buead 15g、Liquoric Root 3g;using the method of unsupervised entropy hierarchical clustering,the correlation degree is 8 and the penalty degree is 2,the new prescription of treating bronchial asthma is obtained:1.Ephedra Herb、Heartleaf Houttuynia Herb、Tatarian Aster Root、Common Yam Rhizome、Earthworm.2.Platycodon Root、Dendrobium、Figwort Root、Largehead Atractylodes Rhizome、Epimedium Herb.3.Lalang Grass Rhizome、Pummelo Peel、Chinese Thorowax Root、Malaytea Scurfpea Fruit.4.Divaricate Saposhnikovia Root、Figwort Root、Chinese Thorowax Root、Ephedra Herb、Tatarian Aster Root、Earthworm.5.Pummelo Peel、Great Burdock Achene、Malaytea Scurfpea Fruit、Chinese Angelica、Epimedium Herb.6.Chinese Mag oliavine Fruit、Common Macrocarpium Fruit、Chinese Angelica、Biond Magnolia Flower、Heterophylly Falsestarwort Root、Epimedium Herb、Great Burdock Achene.Study 2:Based on the conclusion of prescription analysis in Study 1,five kinds of traditional Chinese medicines(Ephedra Herb、Common Coltsfoot Flower、Perilla Fruit、Perilla Leaf and Tatarian Aster Root)were extracted for network pharmacology analysis.After the query of TCMSP,a screening mechanism of ob≥ 30%and DL≥0.18 was set up,91 kinds of effective ingredients were obtained,including 23 kinds of ephedra,22 kinds of winter flowers,14 kinds of Perilla There are 16 species of perilla seed,19 species of aster,785 corresponding targets of components,including 210 ephedra,166 winter flowers,99 perilla,126 perilla seed and 184 aster.After standardizing the name of asthma by PubMed platform,target queries are carried out on the platforms of degenet,genecards and TTD,and 451 related targets of asthma are obtained by combining the three databases;compound targets and disease targets are obtained by combining them 53 core targets were obtained from the intersection of points;the network visualization model of core drug component disease targets was constructed by Cytoscpae 3.7.2 software and the topological property analysis was carried out_The core Chinese medicine was ephedra,the core compounds in the top 3 were quercetin,luteolin andβ-sitosterol,and the core targets in the top 3 were HSP90AB1,PIK3CG and BCL2L1 String database,PPI network analysis,the top three core targets are CXCL8,VEGFA,mapkl.The function analysis of the enrichment genes of the genes of interest obtained by CLUEGO was mainly divided into 12 groups,including 141 specific functions;the pathway analysis of the genes of interest was carried out by David tool,and the estrogen signaling pathway,cholinergic synapse and progesterone mediated oocyte were obtained Match(progesterone mediated oocyte maturation)is a key pathway,in which estrogen signaling pathway is the most important.PI3K Akt signaling pathway in this pathway contains 10 genes of concern,which is one of the important pathways to predict the therapeutic mechanism of Zhang Hongchun.Conclusion:Based on Professor Chao’s experience,Professor Zhang Hongchun’s idea of treating bronchial asthma is to give consideration to the etiology of "wind" and "phlegm" of asthma,the pathogenesis of lung hypostasis and airway spasm,the CO treatment of lung hypostasis and spasm in the position of the disease,the CO treatment of lung hypostasis and spleen,the same tone of mother and son,the combination of cold and heat according to the disease medication,and the combination of pungent and bitter,so that the lung hypostasis is appropriate,and the propaganda of lung hypostasis is not too much damage Lung qi,descending Qi,rising hair and eliminating phlegm,lung recovery and purging,and airway contracture have unique characteristics.In terms of network pharmacology,quercetin,luteolin,β-sitosterol,gene target CXCL8,VEGFA,mapkl;estrogen signaling pathway,cholinergic synapse and progesterone mediated oocyte Progesterone mediated oocyte maturation is more critical,and PI3K Akt signaling pathway in estrogen signaling pathway is highly correlated with the main pathological
