ObjectiveTo mine molecular markers related to the early diagnosis of breast cancer by using the high-throughput data of the GEO database,and to explore the mode of action of the target gene.Methods1.Through a comprehensive search of the GEO database,three optimal data sets GSE29431,GSE21422,and GSE22544 were obtained.2.After the data sets are analyzed by GEO2 R,three sets of differentially expressed genes are screened out,and the common differentially expressed genes of the three sets of data sets are obtained through common word analysis.3.Perform GO biological function enrichment analysis and KEGG signal pathway enrichment analysis of the common differentially expressed genes to obtain the most meaningful pathways for these genes to play a biological role in the organism,and to obtain the gene location,biological function and Participate in the biological process of action.4.Construct a protein interaction network diagram with genes in biologically significant pathways and observe their interactions.5.Perform survival analysis of differential genes through Kalpan-meier plotter,remove genes that are not related to the prognosis of breast cancer,and divide the remaining genes into groups with high expression that have an adverse effect on the prognosis of breast cancer,and high expression has a positive effect on breast cancer.Action group.6.Through literature search,screen the genes of the group with high expression that have adverse effects on the prognosis of breast cancer,extract genes that have not been associated with breast cancer,and introduce such genes into UALCAN to analyze their expression.And for the second verification of survival analysis,the target gene CKS2 was obtained.7.The GEPIA database analyzes the expression of CKS2 in various cancers.8.Query the expression of CKS2 in breast cancer tissue in Human Protein Atlas.9.Checking the literature shows that the expression of CKS2 is related to CDK protein,so the correlation analysis was performed in GEPIA.10.Verify the differential expression of CKS2 in breast cancer tissues and adjacent tissues by immunohistochemistry.Results1.There are 748 differential genes that have intersections in the three data sets,of which 643 genes are up-regulated in breast cancer and 105 genes are down-regulated genes.2.KEGG analysis shows that the differential genes mainly play a role through the PPAR signaling pathway,morphine addiction,AMPK signaling pathway and other signaling pathways.3.GO enrichment analysis shows that the differential genes are mainly located in exosomes,lipid particles,etc.,involved in angiogenesis,brown adipocyte differentiation and other biological processes,playing the role of "binding peptide hormones,transcription activator activity",etc..4.Survival analysis and literature screening show that the high expression of 9 genes(CDC54,CCNB2,HFARP,CKS2,CIB2,PPP1R1 A,DGAT2,ABHD5,HIP3)and the overall poor survival rate are significant5.UALCAN confirms that 4 of the above 9 genes are up-regulated and 5genes are down-regulated in breast cancer.However,survival analysis verified that only the expression of CKS2 was significantly related to the survival rate of breast cancer.6.CKS2 is highly expressed in many malignant tumors.7.CKS2 is also highly expressed in breast cancer tissues.8.The Pearson correlation analysis of CDK and CKS2 in GEPIA shows that CDK10 has the strongest correlation with CKS2(the correlation coefficient is the largest)9.The results of immunohistochemistry showed that the expression difference of CKS2 in breast cancer and adjacent tissues was statistically significant.10.The results of immunohistochemistry showed that the expression of CKS2 has no significant correlation with the annual tumor size,WHO grade,and whether there are tumor thrombi in the vessels of breast cancer patients.ConclusionsCKS2 is highly expressed in breast cancer and reduces the survival rate of breast cancer patients.Its main mode of action is to coordinate the cell cycle process by binding to CDK protein.These two proteins can be used as key molecules to improve the early diagnosis of breast cancer.
