| In this paper,aerobic and anaerobic microbial community structures and functions of green alga Enteromorpha prolifera-degrading consortia were studied.The fementation products were exploited to produce fruits preservatives and biogas.The microflora D2-1 of aerobic digestion and the microflora EM-D of anaerobic digestion were obtained in preliminary work.To value the characteristic of E.prolifera,the compositions of five type algaes from Fujian was analyzed.It turned out that E.prolifera was a nutrient-rich material which contained 36.11%mainly nutrient content(crude protein of 25.88%,crude polysaccharide 9.53%,crude fat of 0.7%).The content of TS,crude fibre,ash,organic carbon,organic nitrogen and carbon nitrogen ratio of E.prolifera was 84.99%,5.20%,5.44%,26.92%,3.50%,and 6.50,respectively.A preservative of E.prolifera was designed by the metabolites of E.prolifera aerobic fermentation for 7 days.The results indicated that the 0.5 times of E.prolifera fermentation liquid had a strong preservation effect on peach during 12 days of storage period.The effects of the preservative contained keeping good fruit rate,hardness and reducing respiratory rate,decay index susceptible index and color difference as well as maintaining the sensory quality of peach.In the study of E.prolifera aerobic fermentation,the variations of TS,crude polysaccharides and organic nitrogen were analyzed.The results appear as shown below:(a)the TS of substrate from 84.99%drop to 29.69%,(b)the crude polysaccharides content of substrate reduced 69.90%,and the crude polysaccharides of the fermentation broth reached the peak with the 2.11 g·L-1 after fermentating for a day.The variations of microflora was sequenced by high-throughput sequencing technology which showed that Cyclobacteriaceae,Microbulbifer,Novosphingobium,Pseudomonas,Rhizobialeand Hyphomicrobiaceae were the major microorganism.Metabonomics analysis results of aerobic fermentation showed that glutamate,glutamine,alanine,ribose,trehalose made a great contribution to metabolites difference between two microflora samples.Study on the structure and function of E.prolifera anaerobic fermentation bacteria included composition transformation,flora structure and function analysis.Transformation of components during fermentation process showed:The biogas production and methane production of E.prolifera anaerobic fermentation reached 346.81 mL·g-1 and 184.46 mL·g-1,respectively.After degration,the contents of residual organic nitrogen,organic carbon,crude fiber and total solids of substrate was 5.39%,27.63%,3.50%,20.51%,respectively.The variations of microflora showed that S1 genus of Thermotogaceae,Coprothermobacter,Clostridium,Caloramator,Buchnera were dominant bacterial communities in the process of E.prolifera degestion,which produced H2,CO2,acetic acid and other substances to substances methanogenic archaea growth.The analysis of the archaeal community showed that Cenarchaeum,Methanothermobacter,Methanosarcina,Methanomassiliicoccus,Methanosaeta were dominant archaea communities in the reactor.Cenarchaeum,which had a ratio more than 80%,was the most abundance archaea.A comprehensive utilization of E.prolifera was achieved through translated E.prolifera to fruits preservatives and biogas.In such a way,E.Prolifera can be utilizated more adequately which not only relieveing the breakout of green tide lead to marine ecological problems,but also exploit a commercial way to produce fruits preservatives,biogas or other goods.This paper was aimed to provide a reference way to realize the diversification and industrialization way of developing green algae E.prolifera. |
