Carotenoids in Lycium barbarum L,as an important bioactive ingredient,have been studied because of their protective functions of retinitis pigmentosa and cataract prevention.At present,most of the researches focus on the function of Lycium barbarum L pigments and extraction methods,and lack of comprehensive and systematic studies on the carotenoids in Lycium barbarum fruits and the bioaccessibility of major carotenoids in Lycium barbarum L under simulated digestion conditions in vitro.This study based on metabonomics method of carotenoids in Chinese wolfberry in qualitative analysis.Furthermore,the color,texture and microstructure of different treatments and different treatments of Lycium barbarum L were simulated for digestion in vitro,carotenoids in digestive juices biological availability and oxidation resistance of study,what kind of way for consumers to select which treatment method allows the maximum amount of carotenoids to be absorbed in the small intestine,concrete research content as follows:(1)Study on carotenoids in Lycium barbarum L LC-Q-TOF-MS high resolution mass spectrometry was used to analyze the carotenoids in Lycium barbarum L based on metabolomics.The results showed that a total of 8 carotenoids,3 carotene esters and 2 carotene glycosidated derivatives were detected under ESI+and APCI+modes.APCI+mode to detect new xanthine,9-or-9-(cis)zeaxanthin,?-cryptoxanthin,alpha carotenoids,9-or-9-(cis)-beta carotenoids,?-cryptoxanthin monopalmitate.Two carotene glycosidized derivatives,zeaxanthin glucoglyceride and 1'-hydroxy-?-carotenoid glucoglyceride,were detected in ESI+mode.And the rest of the new xanthine,13-or-13-(cis)zeaxanthin,total-trans-zeaxanthin,total trans-beta carotene,zeaxanthin single palmitate and corn Huang Sushuang palmitate in APCI+and ESI+common detection mode.Among them,all-trans carotenoids are natural carotenoids in Lycium barbarum L,and their cis structure is produced under certain conditions.In addition,the carotene of Lycium barbarum L were extracted according to the precise molecular weight of ion spectrum and deconvolution operation.After deletes the background noise and the non-repeatable ions in the QC sample,a total of 10,000 ions were detected in the positive ion mode of APCI,and a total of 1484 ions were detected by removing the background noise.PCA score chart was analyzed by principal component analysis.PLS-DA diagram showed that there were significant differences in compounds between Lycium barbarum L and Lycium barbarum L,including not only differences in carotenoids.(2)The color,texture and microstructure of dried,soaked in water and soaked in wine treatment of Lycium barbarum L were analyzed,and the types and bioaccessibility of carotenoids in the digestive juice of three different treatment methods of Lycium barbarum L were simulated by in vitro digestion.The results showed that there was no significant difference between the L*value and the b*value of dried and soaked Lycium barbarum L.The color characteristic value of dried Lycium barbarum Lwas the highest,followed by that of soaked Lycium barbarum L in water and soaked in wine.There were significant differences in texture parameters among three different treatments.The microstructure of dried Lycium barbarum L was destroyed most seriously.Secondly,soaking wine was more serious than soaking wolfberry in water,which was more conducive to the release of carotenoids.The bioaccessibility of carotenoids in three different treatments of Lycium barbarum L was compared as dry Lycium barbarum L>soaked in wine>soaked in water,and zeaxanthin dipalmitate(36.01%)>zeaxanthin(15.0%)>?-carotenoid(0.78%)in dry Lycium barbarum L.(3)The antioxidant activity of carotenoids in the digestive juices of Lycium barbarum L treated in different ways was studied.Because the carotenoids in Lycium barbarum L had antioxidant activity,the antioxidant strength of carotenoids in the digestive juices needed to be further studied,and in this study,the total carotenoid content and bioaccessibility in digestive juice were analyzed with and without corn oil,and the results in Chapter 3 were verified.The results showed that the antioxidant activity in intestinal digestible was as follows:DPPH scavenging ability,FRAP iron reduction ability and ABTS radical scavenging ability were improved by adding corn oil,and the antioxidant activity of carotenoids in intestinal digestible was improved by adding corn oil.In the determination of total carotenoids in digestive juice,the dried Lycium barbarum L in small intestine stage was 1.512 ug/10 mg,that of Lycium barbarum L soaked in wine was 0.767 ug/10mg,and that of Lycium barbarum L soaked in water was 0.327 ug/10mg.And three different treatment of medlar total carotenoids biological availability,dried wolfberry 59.2%respectively,wolfberry wine 48.91%,wolfberry bubble water 43.25%.The bioaccessibility analysis of three different treatments at different pH stages showed that the bioaccessibility of carotenoids at pH=7 was higher than that at pH=2.5.
