With the rapid development of modern society,the pace of our life has been gradually accelerated.The increasing mental pressure,the irregular work and rest time and the lack of longterm physical exercise make people more easily tired,and with the decline of immunity and other problems.Long term exposure to this type of sub-optimal health is associated with a range of diseases.In order to solve the above problems,this paper selects Polygonatum sibiricum,Astragalus membranaceus,Panax quinquefolium,fermented Cordyceps Militaris powder(Paecilomyces batmannii powder)and Wapiti antler to form a compound prescription through the formula design.A safe and stable compound chinese medicine health oral liquid was developed by optimizing the technology,and its anti-fatigue and enhancing immunity effects were studied.1.By searching and analyzing the approved anti-fatigue and immunity dietary supplement,and combining with the dietary supplement list of raw materials,the factors such as the nature,flavor,function,indications,compatibility,safe dosage and cost of the medicinal materials were taken into account The main composition of the compound chinese medicine health oral liquid was determined as follows: Polygonatum sibiricum 6g,Astragalus membranaceus 5g,Panax quinquefolium 2g,fermented Cordyceps Militaris powder(Paecilomyces batmannii powder)1g,Wapiti antle 1g.2.By using the direct water extraction method,the extraction rate of crude polysaccharide,total saponin and adenosine was taken as the index,and the extraction times,extraction time and ratio of material to liquid were optimized by orthogonal test on the basis of single factor study.The optimum process was as follows: extraction times were 3 times,extraction time was 2 hours,and the ratio of solvent to material was 10:1.3.According to the optimized technological conditions,three batches of oral liquid samples were made,and the sensory,routine physical and chemical indexes,symbolic components and microorganisms were detected in 0 month,3 month,6 month,9 month and 12 month.The results show that the process is feasible and suitable for scale-up production,and the oral liquid is stable in12 months shelf life4.The acute oral toxicity test and 30-day feeding test in rats and mice were used to study the safety of oral liquid.In the acute oral toxicity test,there were no obvious behavioral or mental abnormality,no poisoning or death,and the maximum tolerable dose was over 40.0 g/ kg body weight in both rats and mice There was no significant difference in body weight,hematological index,blood biochemical index and SPLANCHNIC body ratio between the three dose groups and the negative control group.The results showed that the oral liquid was non-toxic in the dose range of this test5.The effects of oral liquid on anti-fatigue and enhancing immunity were studied through animal experiments.The results of mice weight-bearing swimming test showed that the oral liquid could prolong the time of mice weight-bearing swimming and reduce the content of blood urea nitrogen and blood lactic acid after exercise,which indicated that the oral liquid had good antifatigue effect The results of cell and humoral immunity and macrophage function of mice were positive,which indicated that the oral liquid had the effect of enhancing immunity.
