Alzheimer’s disease(AD)seriously affects human health,while monoamine oxidase B(MAO-B)is a major target for the treatment of AD.Therefore,research and development of MAO-B inhibitors have always been a hot research topic,and the extraction and isolation of natural active ingredients from medicinal plants is one of the important ways of drug research and development.In previous studies,we isolated a novel coumarin,anisucoumaramide,from Rutaceae plants,and found that the coumarin showed inhibitory activity on MAO-B,with IC50 value of 143.65±0.90 n M。We want to continue to isolate coumarins with novel structures and similar activities.In this paper,a series of molecular docking methods are used to predict that coumarins in Murraya have great inhibitory potential on MAO-B.Therefore,the chemical constituents of Murraya exotica from Yunnan were studied in this paper and 32 coumarins were isolated from them,including a new coumarin exoticatin A(1).The molecular docking of separated coumarins showed that most of the coumarins had good docking results.The interaction of the new coumarin exoticatin A(1)with the positive control anisucoumaramide was very similar,and the score was high.In this thesis,Murraya exotica from Hekou,Yunnan Province was invoked as raw material.After extraction and partition,various separation methods such as silica gel column chromatography and Sephadex LH-20 gel column chromatography are used,combined with nuclear magnetic resonance(NMR),mass spectroscopy(MS)and ultraviolet spectroscopy(UV),co-separation from ethyl acetate extraction part,fifty compounds(1~50)were identified.Among them,there is one new compound,namely exoticatin A(1),the compound belongs to the heterodimer of isopentenyl coumarin and cinnamic acid derivatives.One new natural product,namely ethyl 3-hydroxy-3-(4-hydroxy-3-methoxyphenyl)propanoate(50).48 known compounds(2~49),including 31coumarin compounds,namely exotimarin G(2),trans-dehydroosthol(3),osthol(4),meranzin(5),phebalosin(6),hainanmurpanin(7),murralonginal(8),isomurralonginol acetate(9),isomeranzin(10),paniculonol isovalerate(11),7-methoxy-8-(2-formyl-2-methylpropyl)coumarin(12),muralatin O(13)),isomurranganon Senecioate(14),2′-O-ethylmurrangatin(15),muralatin K(16),yuehgesin-C(17),auraptenol(18),murranganon(19),scopoletin(20),murrangatin acetate(21)),albiflorin-3(22),murraol(23),isomurralonginol(24),isomurralonginol nicotinate(25),2′-acetoxy-3′-dihydroxyl-osthole(26),muralatin P(27),murrangatin(28),meranzin hydrate(29),minumicrolin(30),pranferin(31),murradimerin A(32);seven flavonoids,namely 5,6,7,8,3′,4′,5′-heptamethoxyflavone(33),5-hydroxy-3,7,8,3′,4′,5′-hexamethylflavone(34),3,5,6,7,3′,4′,5′-heptamethoxyilavonol(35),3,5,7,3′,4′,5′-hexamethoxyflavone(36),5,6,7,3′,4′,5′-hexamethoxyflavone(37),sinesetin(38),3,5,7,8,3′,4′,5′-heptamethoxyflavone(39);one sesquiterpene,namely aromadendrane-4β,10β-diol(40);two lignans,namely lariciresinol(41),(-)-berchemol(42);three phenylpropanoids,namely trans-ethylferulate(43),trans-p-coumaric acid(44),isoferulic acid(45);four benzene ring derivatives,namely vanillin(46),4-hydroxybenzaldehyde(47),vanillic acid(48),bis(2-ethylhexyl)phthalate(49).Among them,compounds 22,26,40,41,42and 63 were isolated from the genus Murraya for the first time,and compounds 13,16,and 27 were isolated from Murraya exotica for the first time.
