| Objective : To explore the effect of autologous chondrocyte transplantation combined with microfracture in rabbit articular cartilage regeneration in vitro.Methods:Chondrocytes of knee joint of New Zealand rabbits aged 3-4 weeks were isolated and cultured for 3 generations.New Zealand rabbits aged 3-4 months were used as experimental animals to make cartilage defect models with a depth of 1 mm and a diameter of 3-4 mm at the trochlea of the knee joint.Three micro-fracture holes were made in the cartilage defect area with No.21 injection needle to obtain blood clots after microfracture operation.Micromass culture was carried out in vitro.The experimental group consisted of autologous chondrocyte plus blood clot after microfracture.The control group consisted of blood clot after microfracture.The regenerated cartilage was cultured in vitro for 3 weeks.The performance of regenerated cartilage was evaluated by detecting specific cartilage protein,related growth factors and specific cartilage genes..Results:After 3 weeks in vitro,the microaggregates in the experimental groupwere generally were better than those in the control group.The microaggregates in the experimental group were larger than those in the control group.The expression of chondroitization-related genes in the experimental group was better than that in the control group.WB showed that the related proteins in the experimental group were better than those in the control group.Safranine O staining and Collage II staining showed the secretion of extracellular matrix and the expression of cartilage The mechanical properties of the microaggregates in the experimental group were better than those in the control group..Conclusions:The effect of autologous chondrocyte transplantation combined with micro-fracture on cartilage regeneration in vitro of rabbit joint is better than that of simple microfracture.. |
