Objective:To explore the relationship between single nucleotide polymorphism(SNP)of SH2B1 gene and genetic susceptibility to type 2 diabetes mellitus(T2DM).Methods:The study included 120 T2DM patients(DM group)and 100 healthy controls(NC group)in Shanxi.The fasting blood glucose(FPG),total cholesterol(CHO),triglycerides(TG),Low-density lipoprotein cholesterol(LDL-C),high-density lipoprotein cholesterol(HDL-C),fasting insulin(Fins),glycosylated hemoglobin(Hb A1c);HOMA calculator software calculates the steady-state model (HOMA2).Measure the exon 9 sequence of SH2B1 gene by sanger sequencing method,use SNP analysis software to analyze,find the SNP sites related to T2DM,compare and analyze the relationship between each genotype and T2DM genetic susceptibility,to analyze the SH2B1 gene correlation between polymorphism and T2DM.SPSS25.0 statistical software was used for data analysis,and the analysis of gene polymorphism and T2DM susceptibility was performed by binary logistic regression analysis.P<0.05 indicates that the difference is statistically significant.Results:1.The comparison of clinical data and biochemical indicators between the two groups showed that the BMI,waist circumference,TG,Hb A1 c,FPG,Fins,HOMA2-IR,HOMA2%S and HOMA2%B of the DM group were significantly different from those of the NC group(P<0.001).HDL-C is lower than NC group,the difference is statistically significant(P=0.023).2.Analysis of sequencing results showed that three SNPs gene(rs117374029,rs13288569332,and rs1815578610)were found on the exon 9 of the SH2B13.The difference between the genotypes GG and GT of rs117374029 was statistically significant(P=0.002).The difference between allele G and T groups was statistically significant(P=0.036).The genotypes GG and GA of rs13288569332 had statistically significant differences between the groups(P=0.04),while the G and A alleles had no statistically significant differences.The genotype of rs1815578610 had no significant difference between CC and CG groups(P=0.111),and the difference between alleles C and G was not statistically significant.4.In rs117374029,the risk of T2DM with genotype GT was 12.6 times higher than that of GG type(OR=12.653,95%CI 2.542,62.965).rs13288569332 and rs1815578610 are not significantly related to T2DM genetic susceptibility.5.There are significant differences between rs117374029 genotype GT and GG in HOMA2-IR and HOMA2%S(P<0.001).There are differences in CHO and Hb A1c(P<0.05).Conclusion:Our experiments show that the rs117374029 polymorphism of the SH2B1 gene locus is significantly related to the genetic susceptibility of T2DM,and the genotype GT and GG have significant differences in insulin resistance and insulin sensitivity.
