Objective:To investigate the significance of Syndecan-1 in the pathogenesis of pulmonary fibrosis through clinical and animal experiments.Methods:1.Clinical observation:The serum expression levels of Syndecan-1 and transforming growth factor-β1(TGF-β1)in idiopathic pulmonary fibrosis(IPF)patients and healthy controls were measured by enzyme linked immunosorbent assay(ELISA).Clinical information of the two groups were collected.Correlation between Syndecan-1,TGF-β1 and clinical data(gender,age,WBC,CRP,PO2,PCO2,pulmonary fibrosis index and chest HRCT score)were statistically analyzed.2.Animal experiments:A total of thirty male SD rats were split randomly into control group(group A),2-weeks model group(group B)and 4-weeks model group(group C).The model groups(group B and group C)were induced by bleomycin to build the pulmonary fibrosis rat model.The expression levels of Syndecan-1 and TGF-β1 in the rat bronchoalveolar lavage fluid(BALF)of each group were detected by ELISA.The expression of mi R-29b in rat pulmonary tissue was tested by real-time fluorescent quantitative PCR.Statistical analysis and comparison were performed.Results:1.Clinical observation:Compared with the healthy control group,the levels of serum Syndecan-1 and TGF-β1 in IPF patients were significantly increased,and correlation analysis showed that Syndecan-1 was positively correlated with TGF-β1.The level of Syndecan-1、TGF-β1 in the serum of IPF patients was positively associated with pulmonary fibrosis indexes and HRCT score,and negatively correlated with PO2.2.Animal experiments:(1)The levels of Syndecan-1 and TGF-β1 in the BALF of group B and group C were significantly higher than those of group A;compared with group B and group C,the level of TGF-β1 in group C was significantly higher;but there was no sharp distinction in the level of Syndecan-1 between groups B and C.(2)Compared with group A and group B,the expression of mi R-29b in group C was reduced.(3)In group C,the expression of mi R-29b was negatively correlated with Syndecan-1 and TGF-β1;Syndecan-1 was positively correlated with TGF-β1.Conclusion:1.In the serum of IPF patients and the BALF of rats with pulmonary fibrosis,the expression levels of Syndecan-1 were significantly increased,and the level of Syndecan-1 was positively correlated with the severity of pulmonary fibrosis and the level of TGF-β1.This suggests that Syndecan-1 is an important fibrotic factor.2.The expression level of mi R-29b in lung tissues of pulmonary fibrosis rats was significantly decreased,and was negatively correlated with Syndecan-1.These results suggest that mi R-29b may be involved in regulating the expression of Syndecan-1 in the pathogenesis and development of pulmonary fibrosis,and its mechanism needs to be further studied.
