| purpose of research:Postmenopausal osteoporosis fracture is a more serious problem in postmenopausal women in China,and the operative reduction healing due to the basis of rarefaction of bone,this study through the observation of Migufang(MGF)combined with mechanical vibration intervention to ovarian osteoporosis fracture model rats,aimed to alleviate postmenopausal osteoporosis caused by bone metabolism disorder and promote fracture healing,and to explore the intervention effect of traditional Chinese medicine on internal and external mechanical vibration.research technique:Forty-five SPF Wistar female rats were randomly fed for one week and numbered.Then they were divided into three groups according to the random number table,with 15 rats in each group,including ovariectomized fracture group(OVX),Sham ovariectomized fracturegroup(Sham)and and MGF combined with mechanical vibration intervention group(OVX-MV).The osteopphobic model was established by using the method of removing bilateral ovaries of rats.Bone marrow cavity fixation with Kirschner wire was used to prepare fracture models.Intervention method:(1)MGF intervention: OVX-MV group,on the second day after the fracture operation,the prepared Traditional Chinese medicine decoction of MGF Decoction was given intragastric administration of 100mg/(kg·d).Sham group and OVX group were given the same dose of normal saline,once a day,continuously.(2)Mechanical vibration intervention:OVX-MV group,five days after fracture surgery,the rats were subjected to whole-body vibration with a frequency of 35 Hz,a vibration amplitude of 2mm and an acceleration of 0.5g.Each vibration time was 20 min and the vibration was carried out for 5 days a week.The sham group and OVX group were placed on the vibration table for20 min 5 days after surgery without vibration.Observation indicators and measurement methods:(1)The serum inflammatory factor levels of TNF and Tracp in weeks 2,4 and 6were determined by double antibody sandwich ELISA method.(2)Western blot tests examined the expression levels of FGF23 protein and Sema3 a protein at weeks 2,4,and 6 in rat bone tissue.Result:1.Effect of MGF combined mechanical vibration intervention on serum TNF-expression in rat: the MGF combined mechanical vibration group was significantly reduced compared with ovariectomized fracture group and sham ovariectomized fracture group in the same time period(P <0.01).2.Effect of MGF combined with mechanical vibration on serum Tracp expression in rats:the serum Tracp concentrations of ovariectomized fracture group,sham ovariectomized fracture group and MGF combined with mechanical vibration group were significantly increased in each same time period(P < 0.01).Compared with ovariectomized fracture group,sham ovariectomized fracture group significantly decreased at week 6(P < 0.01).3.Effect of MGF combined with mechanical vibration on the expression of FGF23 in rat bone tissue: At the rat fracture of 2,4 and 6 weeks,the FGF23 in both the sham ovariectomized fracture group and the rat bone tissue were significantly lower than in the ovariectomized fracture group(P <0.01).At weeks 2 and 4,the dense bone square combined with the mechanical vibration group was significantly higher compared with the sham ovariectomized fracture group(P <0.01).4.Effect of MGF combined with mechanical vibration on Sema3 a expression in rat bone tissue: at weeks 2,4 and 6 weeks of fracture,the dense bone combined mechanical vibration group was significantly lower than the sham ovariectomized fracture group(P <0.01);but the expression of sema3 a in the rat bone tissue of MGF combined mechanical vibration group was significantly higher than the ovariectomized fracture group.Conclusion:Combined with mechanical vibration,MGF inhibited the expression of TNF-α and Tracp,alleviated the inflammatory effect,inhibited the differentiation of osteoclasts,improved bone metabolism and promoted fracture healing.It can reduce the expression of FGF23 in bone tissue and up-regulate the concentration of Sema3 a in bone tissue,so as to enhance the activity of osteoblasts and weaken the activity of osteoclasts,thus making bone formation greater than bone absorption and promoting fracture healing after osteoporosis.MGF combined with mechanical vibration intervention can effectively promote the healing of postmenopausal osteoporosis fractures,MGF for internal treatment and mechanical vibration for external treatment,which is an intervention method of integrated traditional Chinese and Western medicine,in response to the call of healthy China and the development of integrated traditional Chinese and Western medicine advantages. |
