Study On The Mir-126 In Angiogenesis Of The Breast Carcinoma

Micro-rna is a non-coding small rna molecule,it play an important role in the cancinogenesis.Studies showed that the mir-126 also related with gastric cancer,lung cancer,breast cancer and so on,Inorde to explore the function of mir-126 in the development process of breast cancer,provide new targets for breast cancer diagnosis,treatment and prognosis.Methods(1)mir-126 expressionwasdetectedin48 casesof invasive ductal carcinoma and 20 cases of benign breast lesions by using RTQ-PCR.(2)mir-126 stably over –expressed MCF7 cells were established by lentiviral.(3)The cellular functional experiment was carried out in mir-126 over-expression cells.The proliferation,migration,invasion of cells were detected through MTTtest,thewoundhealing testand transwelltest,respectively.Results(1)The lentiviral infected MCF7 cells expressed mir-126 2.8 higher than the control group(p<0.05)(2)MTT experiment shoued mir-126 overexpressed cells proliferation significantly decrease during 72 h and 96 h.Wound healing test and Transwell result indicater the inhibitory effect of mir-126 Incellura migration and invasion.Flow cytometry result showed mir-126 would increase the proportion of G1 phase and decrease the proportion.Conclusion(1)The expression rate of mir-126 and VEGF in breast cancer was significantly lower than that of benign lesions(P<0.05)(2)mir-126 lentiviral transfected stably over expressed MCF7 cells were established.(3)mir-126 over–expression inhibited the proliferation,migration and invasion of MCf7 cells and promoted its apoptosis.
