The Anti-tumor Effect And Its Effect On VEGFR-3 And LYVE-1 Expressions Of Guben Yiai Recipe In Lewis Lung Cancer

Objective We take the lewis lung cancer mice as research object to observe the effect of Guben Yiai Recipe by contrasting the growing and transferring situations of lung tomour after applying the low,moderate,high dose of such soup,adriacin and saline.By means of immunohistochemical staining,we detect micro lymphatic vessel density and the expression situation of VEGFR-3 and LYVE-1,to explore the mechanism of Guben Yiai Recipe on inhibiting lymph node metastasis in lung cancer for providing the theoretical basis of its clinical application.Methods Establishing a mouse model of orthotopic lewis lung,and categorize 45 mice into 5 groups at random:the group with low,moderate,high dose of Guben Yiai Recipe,adriacin and saline.After 21 days,the mice were killed to take the tumor tissue,thymus and spleen,weighing them and calculating the average anti-tumor rate and thymus index and spleen index-Detecting the expression of VEGFR-3,LYVE-1 with immunohistochemical staining and using statistical analysis of the significance of the data.Results 1.Guben Yiai Recipe could reduce the weight and the size of tumor.The inhibitory rates of tumor in lewis lung cancer mice in adriacin groupand high,moderate,low dose of Guben Yiai Recipe were 51.36%,34.07%,30.63%and 12.07%.The inhibitory rates of tumor in high,moderate dose group and adriacin group was different significantly from that of saline group(P<0.05);2.Guben Yiai Recipe could increase the weight of thymus and spleen,and improve the immunity.The thymus index and spleen index in high dose group were the biggest,but there was no significant difference between them and the middle dose group(P>0.05),there were significantly different in low dose group,saline group and adriacin group(P<0.05);3.Adopting immunohistochemical staining todetect and observe the MLVD and LYVE-1 and VEGFR-3 expression in tumor tissue.Compared with saline group,the middle,highdose group of Guben Yiai Recipe,adriacin group could obviously decreasethe expression of LYVE-1 and MLVD and VEGFR-3.(P<0.05),between which adriacin group is strongest,second in high dose group of Guben Yiai Recipe.Conclusion Guben Yiai Recipe has the effect on inhibiting the growth of lung cance,significantly increase the immunity of the mice;Guben Yiai Recipe by the ways of down-regulate the expression levels of VEGFR-3 and LYVE-1 to inhibit the generation of tumor lymphangiogenesis,to inhibit tumor metastasis of lymph node,and finally realize its function in antitumor tumor inhibition.
