| Objective Study the influence of Flavonoids from Tetrastigma hemsleyanum Diels et Gilg on the expression of MDSCs,CD4+T,CD8+T,CD4+T/CD8+T,COX-2,PGE2,Arg-1andiNOS.To explore the immune regulatory function and the possible mechanism on reversion of tumour immune evasion of Flavonoids from Tetrastigma hemsleyanum Diels et Gilg.Methods ①Extracted the Flavonoids from the Tetrastigma hemsleyanum Diels et Gilg,divide it into three groups by the different concentration.②Lung tumor bearing Mice models were established and randomly divided into Flavonoids from Tetrastigma hemsleyanum Diels et Gilg high-dose group,middle-dose group,low-dose group,celecoxib group,saline group and normal group.③24h after inoculated,all groups were began to take gavage except the normal group,once a day for 14 days.The survival status,the weight and the growth of tumor tissues of mice in each group were observed every day.14 days after,the mice were sacrificed and the tumor was removed and the tumor inhibition rate was calculated.The expression of COX-2,PGE2,iNOS and Arg-1 in peripheral blood of mice were detected by Elisa method.The flow cytometry was used to detect the expression of MDSCs,CD4+T and CD8+T cells in the spleen lymphocytes of mice.④ANOVA was used to Conduct Statistical Analysis.Results ①The inhibition rate:Flavonoids from Tetrastigma hemsleyanum Diels et Gilg high-dose group was 44.3%,celecoxib group was 36.3%,middle-dose group was 29.2%low-dose group was 15.4%.②The content of COX-2,Arg-1in peripheral blood:The normal group was the lowest,The saline group was the highest.Flavonoids from Tetrastigma hemsleyanum Diels et Gilg high-dose group<celecoxib group<middle-dose group<low-dose group.high and middle dose group of Flavonoids from Tetrastigma hemsleyanum Diels et Gilg compared with celecoxib group,there were no significant differences.(p>0.05).the rest groups had significant differences compared with each other(p<0.05).③The content of PGE2、iNOS in peripheral blood:The normal group was the lowest,The saline group was the highest.celecoxib group<Flavonoids from Tetrastigma hemsleyanum Diels et Gilg high-dose group<middle-dose group<low-dose group.celecoxib group and Flavonoids from Tetrastigma hemsleyanum Diels et Gilg-middle dose group compared with high dose group for the content of PGE2 there were no significant differences(p>0.05),the rest groups had significant differences compared with each other(p<0.05).high and low dose group of Flavonoids from Tetrastigma hemsleyanum Diels et Gilg compared with middle dose group for the content of iNOS there was no significant differences(p>0.05),the rest groups were all in very significant differences compared with each other(p<0.01).④The content of MDSCs in mice spleen lymphocytes:The normal group was the lowest,The saline group was the highest.Flavonoids from Tetrastigma hemsleyanum Diels et Gilg high-dose group<celecoxib group<middle-dose group<low-dose group.celecoxib group compared with Flavonoids from Tetrastigma hemsleyanum Diels et Gilg middle-dose group,there was no significant differences(p>0.05),the rest groups had significant differences compared with each other(p<0.05).⑤The ratio of CD4+T/CD8+Tcells in mice spleen lymphocytes:The normal group was the highest,The saline group was the lowest.Flavonoids from Tetrastigma hemsleyanum Diels et Gilg high-dose group>celecoxib group>middle-dose group>low-dose group.They all had significant differencs compared with each other(p<0.05).Conclusion ①Flavonoids from Tetrastigma hemsleyanum Diels et Gilg had the power to drive the expression of MDSCs down.The high dose group had the best effect.②The Flavonoids from Tetrastigma hemsleyanum Diels et Gilg can reduce the secretion of COX-2,PGE2,Arg-1,iNOS in peripheral blood may be the mechanism why the Flavonoids from Tetrastigma hemsleyanum Diels et Gilg had the function to reduce T cell immune suppression and the Flavonoids from Tetrastigma hemsleyanum Diels et Gilg high-dose group had the best effect.③The flavonoids from Tetrastigma hemsleyanum Diels et Gilg have anti-tumor effect and the high-dose group had the best effect. |
