Objective To extract the polysaccharide in Tetrastigma hemsleyanum Diels et Gilg,determination of molecular weight and the composition of monosaccharide,then to observe the effects of polysaccharide in Tetrastigma hemsleyanum Diels et Gilg on alcohol liver injury in mice.Methods ①By water extraction and alcohol precipitation method to extract Tetrastigma hemsleyanum Diels et Gilg,clover content of polysaccharide as the index,through orthogonal experiment,the effect of extraction time,extraction times,water ratio,alcohol precipitation concentration of clover polysaccharides extraction effect influence,optimize the extraction condition of polysaccharide in Tetrastigma hemsleyanum Diels et Gilg.②By blood biochemical,ELISA and Wesrten blot assay and by detection of serum glutamic pyruvic transaminase(ALT),and aspartate aminotransferase(AST)level;in liver tissue level of liver homogenate malondialdehyde(MDA),reduced glutathione(GSH)superoxide dismutase(SOD),CYP2E1 and JNK1 protein expression situation to study the protective effect of polysaccharide in Tetrastigma hemsleyanum Diels et Gilg with alcoholic liver injury model in mice.Results ①The optimal extraction conditions were as follows:the extraction time was 2.5h;extract 1 time;the water ratio was 1:25;the alcohol concentration was 70%.②Compared with the blank group,the model group ALT,AST,MDA content was significantly increased(P<0.05),GSH,SOD levels were significantly reduced(P<0.05),the protein of CYP2E1 and JNK1 in liver tissue was significantly increased(P<0.05).Compared with the model group,Tetrastigma hemsleyanum Diels et Gilg polysaccharide high dose group(250mg/kg),ALT,AST,MDA were significantly lower(P<0.05),GSH and SOD content was significantly increased(P<0.05),liver CYP2E1 and JNK1 protein expression were significantly decreased(P<0.05),while the low dose group(125mg/kg)each index had no significant change(P>0.05).Conclusion Using water extraction and alcohol precipitation method to extract the polysaccharides in Tetrastigma hemsleyanum Diels et Gilg,optimize the extraction conditions,the method is simple and has good stability and accuracy.Polysaccharide in Tetrastigma hemsleyanum Diels et Gilg has good impedance function on alcohol liver injury in mice.
