Objective To investigate the regulated effect of acupuncture on amino acid neurotransmitter and it’s metabolites in brain of cerebral ischemia rats by observing the dynamic content change of glutamic acid(Glu),γ-aminobutyric acid(GABA)and lactic acid(Lac)in cerebral ischemia extracellular fluid.To explore the specificities of acupoints by acupuncture different points sharing the same meridian.Methods All of the 80 SD rats were implanted with microdialysis cannula,then selected 5 rats as the blank group randomly,the other rats were established into the model of ischemia of Middle Cerebral Artery Occlusion by modified thread-occlusion method,Postoperative,we use Bederson to calculate neurological score and laser Doppler monitor to supervise the real-time baseline brain blood flow.The score of which were greater or equal to 2 and the cerebral blood flow decreased 70%±10%were deemed successful model.The successful model of 44 rats were randomly divided into three group,16 as the model group,16 as the Fengfu(GV16)group,12 as the Shuigou(GV26)group.The rats of GV16 group and GV26 group were acupunc-tured on the point of GV16and GV26 respectively and lasting 30min after balance for 1h.At the same time we collect the basic dialysate at the rate of 2μl/min for 30min with continuously three days by microdialysis techniques,so as to the blank group and model group.The High Performance Liquid Chromatography with Fraunhofer Line Discriminator or Ultraviolet are used to detect the dynamic content changes of Glu、GABA and Lac which are in the extracellular of the focal ischemic brain tissue.Results(1)The result of the Glu showed:when compared with the blank group,the content of the Glu in Dialysate of the model group rats at different points were significantly increased(P<0.05);when compared with the model group,the Glu of GV16 group and GV26 group were decreased(P<0.05),and GV26 group were lower than GV16 group(P<0.05);(2)The result of the GABA showed:when compared with the blank group in the first day,the content of the GABA in Dialysate were increased of the model group(P<0.05);when compared with the model group,GV16 group and GV26 group were no difference;in the second day,when compared with the model group,the GABA were increased of GV26 group(P<0.05)and there was no difference with GV16 group:in the third day,when compared with the model group,the GABA were both increased of GV16 group and GV26 group(P<0.05),and the GV26 group were higher than GV16 group(P<0.05);(3)The result of the Lac showed:when compared with the blank group,the content of Lac in Dialysate of the model group rats at different points were significantly increased(P<0.05);when compared with the model group,the content of the Lac at different points of GV16 group and GV26 group were decreased(P<0.05),and the GV26 group were lower than GV16 group(P<0.05).Conclusions(1)Acupuncture GV16 and GV26 can effectively reduce the content of Glu,Lac and maintain the level of GABA in corpus striatum extracellular fluid of cerebral ischemia rats,and by regulating the content of amino acid neurotransmitter and it,s metabolite in the area of focal cerebral ischemia can improve the early ischemia-induced neuronal injury;(2)The regulating effect on Glu,Lac and GABA in corpus striatum extracellular fluid of cerebral ischemia rats by acupuncture GV26 is better than GV16;(3)GV16 and GV26 which share the same meridian have specific effect on regulating ischemia-induced neurotransmitter metabolic disorder.
