ObjectiveTo investigate the chemical constituents of Chromolaena odorata(L.)R.M.King&H.Rob;To screen the antitumor activity and antioxidant activity of isolated compounds from Chromolaena odorata(L.)in vitro.Methods(1)Isolation and identification of chemical constituentsThe chemical constituents were isolated and purificated by silica gel column,macroporous resin column,Sephadex LH-20 column,ODS column,semi preparative liquid chromatography and recrystallization method.The structure of chemical constituents were identified on the basis of physicochemical properties and spectral analyses.(2)Antitumor activity screening of chemical compoundsMTT was used to screen the antitumor activity.(3)Antioxidant activity screening of chemical compoundsDPPH was used to screen the antioxidant activity.Results(1)Isolation and identification of chemical constituents27 constituents were obtained from the plant,and identified as:naringenin-7,4’-di-methyl ether(1),2’-hydroxy-3,4,4’,5’,6’-Pentamethoxychalcone(2),5,6,7,4’-tetramethoxyflavanone(3),taxifolin-7,4’-dimethylether(4),persicogenin(5),radicol(6),5,6,7,4’-tetramethoxyflavones(7),odoratin(8),stigmasterol(9),5,7-dihydroxy-6,4’-dimethoxyflavone(10),quercetin-7,4’dimethyl ether(11),4,2’-dihydroxy-4’,5’,6’-trimethoxychalcone(12),3,5,4’-trihydroxy-7methoxyflavanone(13),5,7,3’,4’-tetramethoxyflavanone(14),4’-hydroxy-5,6,7-trimethoxy flavanone(15),Sinensetin(16),5,7-dihydroxy-6,4’-dimethoxyflavanone(17),4’-hydroxy5,6,7,3’-tetramethoxyflavone(18),padmatin(19),6-hydroxy-5,7,4’-trimethoxyflavone(20),Chromolaena S(21),3,4’-dihydroxy-5,6,7-trimethoxyflavone(22),taxifolin(23),dihydrokaempferol(24),taxifolin-4’-methyl ether(25),dihydrokaempferide(26),rhamnetin(27).(2)Antitumor activity screening of chemical compoundsThe experimental compounds exhibited a antitumor activity below then 40%.Compound 21 shown a 37.9%antitumor activity in concentration of 100 pg/mL,better than other compounds.(3)Antioxidant activity screening of chemical compoundsCompound 13 shown the best antitoxidant activity,which was about 54.4%in concentration of 5 mg/mL,and compound 21 was 31.3%.Compound 21,23,24,27 shown a capacity of antioxidant activity.ConclusionTwenty-seven compounds were obtained,compound 21 was a novel natural product,compound 6,14,18,20,23-25 were first obtained from the plant,compound 6 was isolated from Chromolaena genus for the first time.The all thriteen compounds shown a weaker activity against HepG2 cells;Compound 13 exhibited a better antioxidant activity than other compounds.Compound 21 has the capacity of antitumor activity and antioxidant activity.
