Expression Of NGF And Its Receptor TrkA,P75 In Epithelial Ovarian Cancer And Its Significance And The Conduction Of NGF SiRNA Expression Vector And Its Significance

Objectives To compare the expression levels of NGF,tyrosinekinasereceptor A(TrkA),p75(NGF receptor,NTR)in epithelial ovarian cancer and normal ovaries.Additionally,to examine the effects on ovarian cancer cell line(A2780)proliferation and apoptosis after NGF down-regulation induced by lentivirus induced RNAi.Methods The expression of NGF,TrkA,and p75 in 19 normal ovarian samples and in 29 epithelial ovarian cancer samples were analyzed by Q-RT-PCR and immunohistochemistry.NGF knockdown in A2780 was achieved by shRNA and proliferation,invasion and apoptosis of cells were examined by MTT,transwell assay and flow cytometry,respectively.The protein levels were examined by Western BlotResults Our results showed that the mRNA expression of NGF,TrkA,p75 in experimental group were 7.62±2.55,11.44± 3.54 and 5.34±3.50 While the control group were1.40±2.85,3.75±1.01 and 4.62±1.63.The former two had significant differences in statistics(T values were 7.674,11.024,P values were 0.016,0.008).While the last one had no difference(t=0.812,p=0.214).The medium to strength expression rate of protein of NGF,TrkA,P75 were 37%,82%and 0%.While the control group were 16%,43%and 16%.Three of them were all had statistically significant(x2 values were 11.399,8.587,9.445,P values were 0.003,0.014,0.009).The inhibitory rate of 24h,48h,72h and 96h:were 0.475±0.023,0.541±0.003,0.628±0.009 and 0.737±0.011 in NGF shRNA group,and it obviously inhibited the growth of NC group(0.535±0.011,0.655±0.036,0.698±0.015,0.848±0.012)and normal cells(0.552±0.007,0.733±0.018,0.767±0.018,0.881±0.003)(F values were 28.914,68.323,94.635246.171,p values were<0.01).Compared with the normal cells,the NGF shRNA cells were significantly inhibited about the capacity of invasion.The statistic showed that rate of NGF shRNA cells was 59.0±7.21%,and there was statical difference(f=91.32,p=001).NGF shRNA cells showed an obvious apoptosis which reached 49.59±1.00%.The numbers of NC cells and normal cells were 0.28±0.05%and 0.26±0.06%.It showed a significant difference(f=72.68,p=0.003).The expression of NGF shRNA strains was 40%and another was 27%,It showed no significant difference between the NC group and normal cells(7he F values were 16.018,13.571 and P values were 0.027,0.035).While there was no difference in P75(f=3.341,p=0.327).Conclusions The expression of NGF and TrkA in EOC was significantly higher than normal cells Knockdown of NGF promoted apoptosis and thus inhibited cellular proliferation in A2780 cells.Thus it is of important significance.
