Swertiae herba which belongs to Gentianaceae family is one of the most commonly used herbs for millennium in Traditional Chinese Medicine(TCM).It is quoted in the Chinese Pharmacopoeia and has multiple pharmacodynamic actions including the treatment of influenze or common cold with fever,stomachic,cholagogue and hepatoprotective.As a treasure of traditional medicine culture,Chinese medicine herb has been used in clinical practice for thousand of years.TCM always acts as an indispensable role in prevention and remedy of disease.In our country,many kinds of TCM derive from different source,so the phenomenon of homonym and synonyn is very common.Even the same plants would have totally different constituents due to different factors,such as geography,climate,region and so on.As rare species,Swertiae herba is extremely limited in amount.The Swertiae herba sold in the market now is mostly substitutes,or even a fake,which varies greatly in quality.Therefore,in order to guarantee the quality of this medicne,it is imperative and urgent to carry out scientific quality control on this kind medicine.In this paper,the methodology of scientific quality control for Swertia pseudochinensis has been studied,and the constituted chemicals of swertiae herba also have been analyzed by HPLC-MS.(1)An appropriate HPLC method for simultaneous determination of swertiamarin,gentiopicroside and swertisin in swertiae herba was established.The constitutes of mobile phase were methanol,acetonitrile and sodium dihydrogen phosphate solution(p H was adjusted by 1%phosphoric acid)which volume ratio was 15:9:76.The flow rate was1m L·min-1.The liquid chromatography equipped with COSMOSIL C18 column,and the determination wavelength was 246nm.The injection was 10μL,and the column temperature was 30℃.The analysis method of gentiopicroside was studied and confirmed.The method was proved to be simple and accurate with high reproducibility,sensitivity and selectivity.It was appropriate for the quantitative analysis of Swertiae herba.(2)The HPLC fingerprint method for quality control of Swertiae herba and Pteridophyta was established,and methodolody was also researched.The reagent peaks has not disturbed the detection of samples.The precision,repeatability and stability of this method were well conformed to the standard and can be used as standardization in the quality control of the above two medicines.(3)The chromatographic fingerprint was analyzed by the similarity evaluation system for chromatographic fingerprint of Traditional Chinese medicines.Otherwise,using the SPSS software classified the medicine herbs by the result of Hierarchical clustering analysis and Principle component analysis,and the classification results were consistent well with the similarity of data.(4)As one of the most obvious chromatographic fingerprint technologies,HPLC-MS plays an important role in analyzing components.In this study,11 chemical components have been analyzed,such as swertiamarin,gentiopicroside,mangiferin,swertisin,sweroside,isoorientin,isovitexin,swertianolin,senburiside-Ⅱ,bellidifodin and1-hydroxy-3,7,8-trimethoxyxanthone.It provides a prosperous foundation for the analysis and utilization of Swertiae herba.
