ObjectiveJianpi Tongfu granules is a new traditional Chinese medicine that is being developed and researched by the research group.It is from the clinical use of Jianpi Tongqinfang,which is effective and reliable for more than 10 years.It is used to treat predominant irritable bowel syndrome with constipation(IBS-C).The liver-stagnant and spleen-deficient IBS-C rat model was used to study the changes of gastrointestinal hormones in serum,and to explore the mechanism of Jianpi Tongfu granules in the treatment of IBS-C.Methods(1)Experiments were conducted to investigate the effect of Jianpi Tongfu granules on the progress of small intestine advancing in mice which was nomal or induced by atropine,and the effect of Jianpi Tongfu granule on the small intestine advancing rate in mice by use activated carbon as a marker to evaluate the effect of Jianpi Tongfu granules on intestinal motility in mice.(2)Experiments on the effects of acetic acid-induced writhing in mice were conducted to observe the effect of Jianpi Tongfu granules on the latency of writhing and writhing in mice induced by glacial acetic acid stimulation,and to evaluate the effect of Jianpi Tongyu granules on pain.(3)The intestinal and Jin dryness IBS-C mice model was established by the combination of early mother-infant separation and water-inhibiting and non-fasting,and the first defecation time,number of pellets,fecal weight,and Bristol classification of the mice were observed.The effect of Jianpi Tongfu granules on the efficacy of IBS-C mice was evaluated.(4)IBS-C model of liver stagnation and spleen deficiency was established by combining maternal and infant separation,ice water gavage,drinking water restriction and low-fiber diet,and the defecation-related indicators,intestinal pressure threshold,and sputum were measured in each group.The relevant indicators of the field experiment,the percentage of syrup preference,the level of serum 5-hydroxytryptamine(5-HT),somatostatin(SS),substance P(SP),and the rate of small intestine advancing were investigated to explore the mechanism of Jianpi Tongfu granules in improving the symptoms of liver-stagnation and spleen deficiency type IBS-C rats.Results(1)Effects on the progress of small intestine advancing in normal mice,the results showed that the intestinal propulsion rate in the high,medium,and low dose groups of Jianpi Tongfu granules was significantly increased(P<0.05 or P<0.01).The effect of atropine-induced suppression of intestinal propulsion in mice showed that compared with the normal control group,the intestinal propulsion rate of the model control group mice was significantly reduced(P <0.01).compared with the model control group,the intestinal propulsion rate in the high,medium,and low dose groups of Jianpi Tongfu granules was significantly increased(P<0.05 or P<0.01).(2)The experimental results of writhing induced by glacial acetic acid in mice showed that compared with the control group,the number of writhing in the high and middle dose groups of Jianpi Tongfu granules was significantly reduced(P<0.05 or P<0.01).(3)Effects on the intestinal and Jin dryness IBS-C mice model,the experimental results showed that compared with the normal control group,the number of feces pellets,feces weight,and Bristol classification scores were significantly reduced in the model control group(P<0.01),defecation time significantly prolonged(P<0.01);compared with the model control group,the total fecal pellet count,fecal weight,and Bristol classification scores in the high,middle,and low dose groups of Jianpi Tongfu granules were significantly increased(P<0.05 or P< 0.01),the defecation time was significantly shorter(P<0.05).(4)Effect on the model of IBS-C rat model of liver stagnation and spleen deficiency,the experimental results showed that compared with the normal control group,the number of fecal pellets,fecal moisture content,and Bristol classification scores in the model control group,intestinal pressure threshold,retention time in the central compartment,vertical movement,horizontal movement,number of feces,percentage of sugar water,and intestinal propulsion rate were all significantly reduced(P<0.01),The serum levels of 5-HT,SS,and SP in the serum of rats in the model control group were increased(P<0.01);compared with the model control group,the number of feces pellets,feces moisture,and Bristol classification scores of rats in the high,middle,and low doses of Jianpi Tongfu granules were significantly increased(P< 0.05 or P<0.01).Rats in the high,middle and low doses of Jianpi Tongfu granules had significantly higher intestinal pressure threshold(P<0.01).Rats in the high,middle and low doses of Jianpi Tongfu granules had significantly increased retention time in the central compartment and horizontal movement(P<0.05 or P<0.01).The percentage of percentage of sugar water of rats in the high and medium dose groups of Jianpi Tongfu granules was significantly higher(P<0.05 or P<0.01).The serum levels of 5-HT,SS,and SP in the serum of high,middle,and low dose groups of Jianpi Tongfu granules were significantly lower(P<0.05 or P<0.01).the intestinal propulsion rate in rats of high and medium dose groups of Jianpi Tongfu granules was increased(P<0.05 or P<0.01).ConclusionJianpi Tongfu granules have a role in promoting bowel motility and analgesia,have a better therapeutic effect on IBS-C,and can obviously improve symptoms such as constipation and abdominal pain.Its mechanism of action for treatment of IBS-C with liver stagnation and spleen deficiency may be reduced.5-HT,SS,SP content.
