| Axonal recovery of central nervous system plays a very important role in neurodegenerative treatment.Mammal neurons and axons can not regenerate after destruction.Recovery of central nervous axonal injury becomes one of the significant issues.Rehabilitation includes two aspects: axonal regeneration and neuronal regeneration.Sympathetic neuronal differentiation occurs by nerve growth factor(NGF)sitimulation.It is used as a model of neuronal cell research for the study of nerve cell damage and death of neurotoxicity.Currently,the theory of neurotrophic hypothes is that nerve growth factor can promote the prominent growth and improve neuronal atrophy has been widely accepted.However,the difficult is that nerve growth factor is too lager to go through the blood-brain barrier.Traditional herbal has a wide use in the treatment of central nervous system diseases.The composition and mechanism is too complex to study in the research.This article aims to find potential directions for the treatment of these diseases by studying the proliferation and differentiation of PC12 cells from the simulation of single drugs from Centella and mechanical stimulation.The experimental analysis and data modeling,illustrated that the combination of Asiatic acid,combination of Madecassic acid and Madecassoside,combination of Quercetin and Iisoquercetin,combination of Asiatic acid and Madecassic acid showed a significant role in promoting differentiation.Furthermore,we combined the drug combinations and the shear force in the microfluidic chip.The chip was fabricated by optimizing the hardness of PDMS substrate.The results showed that the increase of substrate hardness was conducive to cell adhesion and proliferation by quantifying the differentiation morphology and cell survival.Finally,two optimized combinations of Centella asiatica(Asiatic acid and Madecassic acid)were selected for intervention.The fluid velocity is 10 μL/h and the shear stress is 0.20~0.23 Pa can enhance the differentiation of PC12 under the action of drug combination when chip size is 4cm * 1500μm * 80μm.It proved that the combination of Centella asiatica could obviously promote the differentiation of PC12 cells compared with single drug,and under the combination of fluid shear force,the differentiation of PC12 cells could be better enhanced.It also provided a new idea for the study of small molecule drugs and fluid mechanics on neuronal cells. |
