Effects Of The Theory Of Eight-periods Chinese Medicine On The TGF-β1、BMP15 Pathway Of The Treatment Effect In IVF Patients With Kidney Yin Deficiency

Objective: To investigate the effects of the theory of eight-periods Chinese medicine on the Expression of genes and proteins of TGF-β1 and BMP15 in ovarian granulosa cells(GC)and on oocyte quality of who had in vitro fertilization of infertile patients with Kidney Yin deficiency.Methods: Eighty infertile patients with Kidney Yin deficiency type treated by in vitro fertilization and embryo transfer(IVF-ET)were divided into eight-periods Chinese medicine treatment group and the control group according to the order of the patients.Patients in both groups were treated with Gn RH-a regimen.The treatment group was treated orally with eight-periods theoretical therapy at the beginning of menorrhagia one month before entering the short regimen of Gn RH-a,and the control group was given the same dose of Liuwei Dihuang Chinese medicine at the same time.The changes of syndrome integral of Kidney Yin deficiency before and after treatment,the control of basic Fsh、Lh、Fsh/Lh、AMH were compared before and after treatment,and the cancellations of cycle,the dosage and days of Gn,the blood level of HCG,the eggs’ number,the fertilization rate,the available and embryo rate were compared between the groups before and after treatment.Pregnancy rate and expression of TGF-β1 and Bmp15.Results:After treatment,the improvement of kidney yin deficiency syndrome in the treatment group was more obvious than the control group,and there was statistical significance between the groups.The level of Fsh was controlled in both groups,but the effect of treatment group on improving Fsh/Lh was significant(p<0.05).The use of Gn in treatment group was less than control group,and in HCG-day’s the treatment group was higher than that in the control group(p<005).The number of eggs,fertilization rate,available embryo rate and cycle clinical pregnancy rate in the treatment group were higher than those in the control group(p<0.05).There was no significant difference in blastocyst rate between them.The results showed that the expression of TGF-β1 and BMP15 was up-regulated in the treatment group and the whole cell level of TGF-β1and BMP15 was increased in the treatment group compared with the control group(P<0.05).Conclusion: 1.The therapy of theory of eight-periods Chinese medicine can obviously improve the state of Kidney Yin deficiency,improve the level of basic FSH,reduce the the dosage and days of Gn,and increase the normal fertilization rate and the rate of available embryos in infertility patients with Kidney Yin deficiency.2.The therapy of theory of eight-periods Chinese medicine may promote the proliferation and differentiation of granulosa cells by activating the expression of TGF-β1 and BMP15,and improve the quality of egg cells and the development potential of embryos.3.Based on the expression of TGF-β1 and BMP15,the theory system of traditional Chinese medicine(TCM)theory of eight-stage theoretical therapy was improved from the level of effector molecule.
