| Objctive:Observation on the Effect of Shizaotang on Idiopathic pulmonary Fibrosis and Its Effect on Expression of VEGF and MMP-9;Study on the Clinical Effect of Shizaotang on Idiopathic pulmonary Fibrosis(IPF);Through Animal Experiments,observe the effect of Shizaotang on the expression of VEGF and MMP-9 in lung tissue of pulmonary fibrosis rat model,explore the mechanism of Shizaotang on IPF,and provide experimental basis for Shizaotang to treat IPF.Methods: Clinical study: Forty-eight IPF patients were selected as the research object,24 cases in the control group and 24 cases in the observation group.The control group was treated with acetylcysteine,and the observation group was treated with Shizaotang on the basis of the control group.The improvement of chest HRCT and lung function scores of patients were observed and compared to evaluate its effectiveness. Animal experiment:96 healthy SD male rats were randomly divided into 6groups: blank group,model group,pirfenidone group,Shizaotang low dose group,Shizaotang medium dose group and Shizaotang high dose group.pulmonary fibrosis model was established by instilling bleomycin through trachea.Intervention therapy was started the next day.Eight rats were killed on the 14 th day and 28 th day respectively.The right lung was harvested for pathological observation and immunohistochemistry was used to detect VEGF expression.The left lung tissue was taken for ELISA to detect MMP-9 expression.The experimental data were statistically processed by SPSS17.0 statisticalsoftware.Results: Clinical research results:(1)chest HRCT scores: There was no significant difference between the control group and the observation group before and after treatment(P>0.05);There was no significant difference in the score between the control group and the observation group after treatment(P>0.05).(2)pulmonary function: FVC(%),TLC(%),DLco(%)in the control group had no significant difference before and after treatment(P>0.05).FVC(%)and TLC(%)in the observation group after treatment were improved compared with those before treatment,with significant difference(P<0.05),and with significant difference compared with the control group(P<0.05).The DLco(%)in the observation group had no significant difference compared with that before treatment(P<0.05). Animal experiment results :(1)pathological results showed that alveolar inflammation appeared on the 14 th day and pulmonary interstitial fibrosis formed on the 28 th day in the model group.The degree of pulmonary fibrosis in each treatment group was less than that in the model group on the 14 th and28th days.(2)ELISA test results:On the 14 th and 28 th days,the expression of MMP-9 in lung tissues of each group was significantly different(P<0.01).the blank group had the lowest expression and the model group had the highest expression.There were significant differences(P<0.01).The expression of MMP-9 in lung tissue of each treatment group was lower than that of the model group,with significant difference(P<0.01).The high dose group of shizaotang is lower than other treatment groups,with significant difference(P<0.05).There was no significant difference between the groups of shizaotang(P>0.05).(3)immunohistochemical results:On the 14 th and 28 th day,the expression of VEGF in lung tissue of each group showed significant difference(P<0.01),the expression in blank group was the lowest,and the expression in model group was the highest,showing significant difference(P<0.01).The expression of VEGF in each treatment group was lower than that in the model group,with significant difference(P<0.01).On the 14 th day,there was no significant difference between the treatment groups(P>0.05).On the 28 th day,the high dose group of shizaotang was lower than other treatment groups,with significant difference(P<0.05).There was no significant difference in the expression of VEGF in lung tissue between the low dose group,middle dose group and pirfenidone group(P>0.05).Conclusion: Shizaotangcan reduce the performance of HRCT inflammatory exudation in patients with partial IPF and improve lung function.Animal experiments prove that Shizaotang can play an anti-fibrosis role by reducing the expression of VEGF and MMP-9 in rat lung tissue. |
