| Objective:Coronary angiography(CAG)is now widely used in the diagnosis of Coronary heart disease.Our previous studies showed that the contrast medium induced apoptosis,inflammation and oxidative damage in human umbilical vein endothelial cells.The purpose of this study was to investigate the effects of iodixanol on RAW264.7 macrophages and its possible mechanism.Methods:Cell experiment:RAW264.7 macrophages were cultured on the standard conditions and incubated with iodixanol injection.The release of reactive oxygen species(ROS)in macrophages was measured by using oxidate probe,Dihydroacetoacetic acid dichlorofluorescent yellow.Transwell cell was used to evaluate the migration ability of RAW264.7 macrophages.Western blotting was used to detect the expression of oxidative stress-related proteins,such as NADPH oxidase activating protein P47,and NF-κB p65.Animal experiment:20 male SD rats were randomly divided into Normal group(PBS)and contrast media group(iodixanol 10ml/kg).Two groups were injected with PBS and iodixanol respectively through the tail vein.Rats were sacrificed,and the kidneys were tested for morphological observation.Results:Iodixanol induced PKC and p65 activation and NOx expression,increased ROS release and promoted cell migration in RAW264.7 macrophages.HE staining in the kidney of rats treated with iodixanol can cause structural disorder,edema,necrosis and other pathological changes of renal interstitial cells.Conclusion:Iodixanol can induce oxidative damage in RAW264.7 macrophages,which may be caused by the activation of PKC and p65 and the expression of NOx.Iodixanol can also promote cell migration in RAW264.7 macrophages,and caused the interstitial cells of kidney edema,necrosis and other changes. |
