ObjectiveTo investigate the effect of IL-8 on the proliferation,migration and invasion of cervical cancer Caski cells,and possible signaling pathway.MethodsCervical cancer Caski cells were cultured in vitro overnight without serum.1、Caski cells were treated with different concentrations of IL-8(0、20、40、60、80、100ng/m L),0ng/m L group serves as control.MTT experiments was used to detect the proliferation of Caski cells.2、The experiment was divided into(1)control group(equal volume DMSO instead of LY294002);(2)IL-8 group;(3)IL-8+LY294002 group;(4)LY294002 group.The concentration of IL-8 was 80ng/m L,the concentration of PI3K/AKT pathway specific inhibitor LY294002 was 20μmol/L.The proliferation of Caski cells were tested by MTT,the migration of Caski cells were tested by scratch test,the invasion of Caski cells were tested by Transwell assay,Western blot was used to analyze AKT,p-AKT,Vimentin,β-catenin and E-cadherin expression.Results1、MTT showed that IL-8 promoted Caski cell proliferation in a dose-dependent manner,and there were no significant difference between80ng/m L and 100ng/m L group(P>0.05),Caski cell proliferation of IL-8 combinedwith the LY294002 group and the LY294002 group were lower than the IL-8group(P<0.01).2、The scratch test and Transwell test showed that Caski cell migration and invasion of the IL-8 group were enhanced(P<0.01).The Caski cell migration and invasion ability of IL-8 combined with the LY294002 group and the LY294002 group were lower than the IL-8 group(P<0.01).3、Western blot showed that there was no significant difference in the expression of total AKT protein in each group(P>0.05).The expression of p-AKT、Vimentin and β-catenin increased in IL-8 group,E-cadherin decreased(P<0.01),AKT pathway specific inhibitor LY294002 can block the effects above mentioned.ConclusionIL-8 may affect the expression of Vimentin,β-catenin and E-cadherin through PI3K/AKT pathway,and promote the proliferation,migration and invasion of cervical cancer Caski cells.
