| Objective:To detect the expression ofβ-catenin,E-cadherin,vimentin and N-cadherin protein in premenopausal and postmenopausal non cervical lesion tissues,LSIL,HSIL and CSCC tissues by immunohistochemistry to explore the pathogenicity of HR-HPV infection in postmenopausal women.Methods:A total of 128 HR-HPV positive cervical tissues in the department of obstetrics and gynecology of the Affiliated Hospital of Inner Mongolia Medical University from December2020 to January 2022 were collected.14 cases of postmenopausal tissue without cervical lesions,15 cases of LSIL,20 cases of HSIL and 15 cases of CSCC confirmed by histopathology were included in the experimental group,and 14 cases of premenopausal tissue without cervical lesions,15 cases of LSIL,20 cases of HSIL and 15 cases of CSCC were included in the control group.Immunohistochemical method(S-P method)was used to detect the expression ofβ-catenin,E-cadherin,vimentin and N-cadherin proteins in each group.Results:1.The positive rates of E-cadherin in HR-HPV positive postmenopausal non cervical lesions group,LSIL group,HSIL group and CSCC group were 92.86%(13/14),73.33%(11/15),35.0%(7/20)and 26.67%(4/15).There was significant difference among the four groups(χ~2=18.22,P<0.001).The positive rates of vimentin in HR-HPV positive postmenopausal non cervical lesions group,LSIL group,HSIL group and CSCC group were14.29%(2/14),20.0%(3/15),55.0%(11/20)and 86.67%(13/15).There was significant difference among the four groups(χ~2=20.43,P<0.001).The positive rates of N-cadherin in HR-HPV positive postmenopausal non cervical lesions group,LSIL group,HSIL group and CSCC group were 7.14%(1/14),13.33%(2/15),60.0%(12/20)and 80.0%(12/15).There were significant differences among the four groups(χ2=23.56,P<0.001).In postmenopausal CSCC,the positive rates of E-cadherin,vimentin and N-cadherin were not related to FIGO stage,pathological grade and vascular tumor thrombus(P>0.05).2.The abnormal expression rates ofβ-catenin in HR-HPV positive postmenopausal non cervical lesions group,LSIL group,HSIL group and CSCC group were 14.29%(2/14)、26.67%(4/15)、65%(13/20)、80%(12/15).There were significant differences in the abnormal expression rates among the four groups(χ2=17.56,P=0.001).In postmenopausal CSCC,the abnormal expression rate ofβ-catenin was not related to FIGO stage,pathological grade and vascular tumor thrombus(P>0.05).In postmenopausal cervical lesions group(including LSIL,HSIL and CSCC),the abnormal expression rate ofβ-catenin was negatively correlated with the positive rate of E-cadherin(r=-0.307,P=0.03),positively correlated with the positive expression rate of vimentin(r=0.353,P=0.012),and positively correlated with the positive expression rate of N-cadherin(r=0.318,P=0.024).3.In the same level of cervical lesions,there was no significant difference in the expression rates ofβ-catenin,E-cadherin,vimentin and N-cadherin between postmenopausal group and premenopausal group(P>0.999).Conclusion:1.In postmenopausal cervical lesions infected with HR-HPV,the positive expression of E-cadherin protein gradually decreased and the positive expression of N-cadherin and vimentin protein gradually increased with the progression of the lesions.Therefore,it is speculated that EMT may be involved in the pathogenicity after HR-HPV infection and the occurrence and progression of postmenopausal cervical lesions.2.In postmenopausal cervical lesions infected with HR-HPV,with the progression of the lesions the abnormal expression rate ofβ-catenin protein increased gradually and was related to the expression level of E-cadherin,vimentin and N-cadherin protein,β-catenin may cooperate with EMT to participate in the pathogenicity after HR-HPV infection,participate in the progression of cervical squamous intraepithelial lesion in postmenopausal women,and promote the occurrence of CSCC. |
