| ObjectiveIn this study,through the systematic pharmacological research methods,the mechanism of Sanhuang granules in the treatment of neonatal jaundice was initially discussed.At the same time,by constructing a rat jaundice model,Sanhuang granules were used for interventional treatment,and the pathological changes and liver pathological changes in rats Serum biochemical indicators were studied to prove the therapeutic effect of Sanhuang Granules on jaundice.Methods1.The Chinese medicine composition database is constructed through important databases such as HIT,BATMAN-TCM and TCMSP.Combined with the analysis of the pharmaceutical composition of the Chinese medicine compound and the manual search database of the research group,the ingredients contained in Yinchen,Dahuang,Huangqin,Huangbai,Gancao are collected.Screening the collected compounds for pharmacokinetic indexes and drug-like index with the help of modern network tools will meet the requirements of oral bioavailability(OB)≥ 30%,drug-like(DL)≥ 0.18,Caco-2 ≥-0.4 As the candidate compounds,the chemical components are screened to obtain active compounds with higher research value.The prediction of the target of the active compound is collected based on the STITCH database,and the target of jaundice is collected by OMIM and Dis Ge Net.2.With the help of Cytoscape software to build a network diagram,three-dimensional representation of the relationship between components,targets and diseases.Analyze from the perspective of the network,and use the degree value and the centrality of the intermediary as indicators to evaluate the importance of components and targets.And through the enrichment analysis of GO biological process and the KEGG pathway enrichment analysis of the targets where the drug and the disease coincide,the preliminary possible mechanism of Sanhuang granules in treating jaundice is revealed.3.Divide SD rats into 5 groups,namely blank control group,model group,positive control group,Sanhuang granule low-dose group,Sanhuang granule middle-dose group,Sanhuang granule high-dose group,except the blank control group,Give ANIT sunflower oil solution(0.6ml / 100g)intragastrically,construct jaundice model,blank control group give equal amount of normal saline,48 h after modeling,positive control group give ursodeoxycholic acid capsule content solution(0.3 mg / 100g)intragastrically,the model group was given the same amount of sodium chloride solution,and the three,yellow and low dose groups were given different concentrations of three yellow granule solutions(70mg / ml,90 mg / ml,110 mg / ml)After intragastric administration,liver samples were collected four weeks later,and the middle lobe of the rat was cut to make pathological sections to observe the pathological changes of the liver.Results1.Proinflammatory factors such as IL6 and TNF may regulate immune function through the NF-k B and MAPK pathways.TNF is a marker of inflammatory response and can eliminate toxic substances through multiple pathways.2.After administration of high-dose,medium-dose and low-dose infusion of Sanhuang granules in rats,the serum liver function index values of SD rats were reduced to varying degrees compared with the model group,confirming that the Sanhuang granules can be reduced by reducing the high Bilirubinemia can achieve the effect of treating jaundice.Observation of pathological slices of jaundice rats showed that the liver fibrosis indexes of rats after Sanhuang granule treatment were significantly improved,and there were also high,medium and low dose groups.difference.ConclusionsSanhuang granules have the characteristics of multiple components,multiple targets,and multiple pathways,which may be regulated by targets such as IL6,TNF,VEGFA,and CASP3,and play the role of turning yellow through the NF-k B and MAPK pathways.Experiments have shown that Sanhuang Granules are effective in medium doses,and can reduce liver pathological damage caused by jaundice disease and reduce liver fibrosis. |
